RP-HPLC测定复方双嘧达莫乳膏中3种主要成分含量
Determination of three main components in compound dipyridamole cream by RP-HPLC
【摘要】 目的 建立测定复方双嘧达莫乳膏中双嘧达莫、盐酸达克罗宁、消旋山莨菪碱3种主要成分含量的方法。方法 采用反相高效液相色谱法。色谱柱为InertSustain C18色谱柱,流动相为甲醇-0.02 mol/L磷酸二氢钾溶液(磷酸调pH值至3.0),流速为1.0 mL/min,检测波长为280 nm和220 nm,柱温为25℃。结果 双嘧达莫、盐酸达克罗宁、消旋山莨菪碱分别在247.4~1 484.4μg/mL、24.2~145.2μg/mL、50.0~250.0μg/mL浓度范围内与其峰面积呈良好的线性关系(r=0.999 4、r=0.999 8、r=0.999 5),加样回收率分别为99.6%、99.5%、99.1%。结论 RP-HPLC方法操作简便,重复性好,结果准确,可用于复方双嘧达莫乳膏的质量控制。
【Abstract】 Objective To establish a method for the determination of dipyridamole, dyclonine hydrochloride and raceanisodamine in compound dipyridamole cream.Methods RP-HPLC method was adopted. The determination was performed on Inert Sustain C18 column with methanol-potassium dihydrogen phosphate(pH was adjusted to 3.0 by phosphoric acid) as mobile phase at the flow rate of 1.0 ml·min-1. The detection wavelength was set at 280 nm and 220 nm, and the column temperature was 25 ℃.Results The linear range was 247.4~1 484.4 μg/ml(r=0.999 4) for dipyridamole, 24.2~145.2 μg/ml(r=0.999 8) for dyclonine hydrochloride, and 50.0-250.0 μg/mL(r=0.999 5) for raceanisodamine. The average recoveries were 99.6%, 99.5%, and 99.1%.Conclusion RP-HPLC method is simple, reproducible, accurate, and can be used for the quality control of compound dipyridamole cream.
