酶法辅助胶体磨同时提取糯米淀粉和蛋白质的工艺研究
Study on simultaneous extraction process of glutinous rice starch and protein by enzyme assisted colloid mill
【摘要】 以糯米为原料,采用酶法辅助胶体磨偶联离心处理同时提取淀粉和蛋白质,对酶法浸泡工艺进行优化。研究结果表明,粗粉碎后的糯米与蒸馏水料液比1:10混合,调节pH值至4.5,加入质量比0.8%的果胶酶,在50 ℃酶解60 min,采用胶体磨碾磨成糯米浆,在4 ℃经6 500 rad/min离心作用后,上清液中蛋白质提取率可达43.8%,滤饼中淀粉得率可达93.2%。方法最大限度保留蛋白质和淀粉分子结构,为后续产业化生产提供一定的参考。
【Abstract】 In this study,the glutinous rice was used to extract starch and protein simultaneously by enzyme assisted colloid mill coupling centrifugation,and the enzymatic soaking process was optimized.The results show that the optimal enzyme assisted process is as follows:mix the coarsely crushed glutinous rice with the distilled water at a ratio of 1:10,then adjust pH value of the mixture to 4.5,and add the pectinase at the mass ratio of 0.8%,and then incubate at 50 ℃ for 60 min for the enzyme hydrolysis.After the pretreated glutinous rice pulp is milled by colloid mill,and after the centrifugation at 6 500 rad/min 4℃,the protein extraction rate of the supernatant can reach 43.8% and the starch yield of the filter cake can reach 93.2%.The method can retain the molecular structure of protein and starch to the maximum extent,which can provide a certain reference for the subsequent industrial production.
