骨碎补总黄酮对牵张成骨模型大鼠中H血管及成血管-成骨耦联的作用
Effects of total flavones of Rhizoma Drynariae on the type of H vessels and angiogenicosteogenic coupling in rat models of distraction osteogenesis
【摘要】 目的:探究牵张成骨(DO)模型大鼠中H血管的表达及骨碎补总黄酮(TFRD)的干预作用。方法:构建DO模型,并构建4 mm骨缺损(BD)模型作为对照,再根据是否应用TFRD干预分为中药和对照两个亚组,即60只SD大鼠分为4组:DO对照组、DO中药组、BD对照组、BD中药组,每组15只。行X线、HE和Masson染色评估成骨情况,血管造影观察血管形成,免疫荧光鉴定H血管表达,免疫组化检测组织内CD31、VEGF、RUNX2和OSX的表达。结果:X线、HE和Masson染色显示,DO模型比BD模型截骨间隙内有更多的骨组织和血管生成(P<0.05),且与对照组比较,中药组中的骨组织和血管生成更多(P<0.05)。与BD模型比较,DO模型中生成更多的H血管(P<0.05),且TFRD可明显促进H血管的表达(P<0.05)。DO模型的CD31、VEGF、RUNX2和OSX表达高于BD模型(P<0.05),且TFRD可显著上调上述因子的表达水平(P<0.05)。结论:H血管存在于DO模型之中,TFRD可协同牵张应力促进H血管生成,增强成血管-成骨耦联作用,继而促进血管生成和骨生成效果。
【Abstract】 Objective: To explore the expression of type H vessels in rat model of distraction osteogenesis(DO) and effects of total flavones of Rhizoma Drynariae on type H vessels. Methods: The DO model was constructed, and the 4 mm bone Defect(BD)model was constructed as the control. Then, according to whether TFRD intervention was used or not, the SD rats in either model were divided into two subgroups. Namely, sixty SD rats were randomly divided into four groups: the DO(CON) group, DO(TFRD)group, BD(CON) group and BD(TFRD) group, 15 rats in each group. X-ray, HE and Masson staining were performed to evaluate the osteogenesis. Angiography was performed to observe the angiogenesis. Immunofluorescence was used to identify the expression of H vessels, and immunohistochemistry was used to detect the expression of CD31, VEGF, RUNX2 and OSX in the tissues. Results: The DO model had more newly-formed bone tissue and angiogenesis than the BD model(P<0.05), and the TFRD group had more new bone tissue and blood vessels than the control group(P<0.05). Compared to BD model, more H vessels were generated in DO model(P<0.05),and TFRD significantly promoted the expression of H vessels(P<0.05). The expressions of CD31, VEGF, RUNX2 and OSX in the DO model were higher than those in the BD model(P<0.05), and TFRD significantly up-regulated the expression levels of these factors(P<0.05). Conclusion: Type H vessels exist in the DO model, and TFRD can promote the formation of type H vessels in coordination with tensile stress, which would enhance angiogenic-osteogenic coupling and subsequently promote angiogenesis and bone formation.
