不同稀释液及稀释比例对关中奶山羊精液低温保存效果的影响
Effects of Different Diluents and Diluting Ratios on the Low-temperature Preservation of Guanzhong Dairy Goat Semen
【摘要】 [目的]试验通过比较精子活率和精子的有效存活时间,探讨在4℃环境下不同稀释液对关中奶山羊精子保存效果的影响,并使用保存效果最好的稀释液进一步研究关中奶山羊精液最佳稀释比例。[方法]试验共设计6种精液稀释液配方,对精液进行1∶4稀释,每间隔6 h检测精子活率并记录精子有效存活时间,筛选出最佳稀释液配方;使用筛选出的稀释液配方对精液进行不同比例的稀释(1∶2、1∶4、1∶8、1∶16和1∶32),每间隔6 h检测精子活率并记录精子有效存活时间,筛选出最佳稀释比例。[结果]以柠檬酸钠作为主要缓冲物质并添加15%卵黄的C组稀释液,在相同保存时间下的精子活率和精子的有效存活时间均显著优于其他5组(P<0.05);使用C组稀释液对精液进行不同比例稀释后,稀释比例为1∶8时在相同保存时间下的精子活率和精子的有效存活时间均显著优于其他组(P<0.05)。[结论]以柠檬酸钠作为主要缓冲物质并添加15%卵黄,稀释比例为1∶8时,精液冷冻保存效果最好。
【Abstract】 [Objective] By comparing the sperm viability and the effective survival time of sperm, this experiment discussed the effects of different diluents on the sperm preservation effect of Guanzhong Dairy Goat at 4 ℃, and the best preserved diluent was used to further study the optimal dilution ratio of semen of Guanzhong dairy goat. [Method] In this experiment, six kinds of semen diluent formulations were designed. The semen was diluted as 1∶4. The sperm viability was detected every 6 hours and the effective survival time of sperm was recorded. The best diluent formulation was selected; the semen was diluted in different proportions with the selected diluent formulation(1∶2, 1∶4, 1∶8, 1∶16 and 1∶32). The sperm viability and effective survival time were measured every 6 hours, and the best dilution ratio was selected. [Result] The sperm viability and effective survival time of group C with sodium citrate as the main buffer and 15% yolk were significantly better than those of the other 5 groups at the same storage time(P<0.05); after diluting semen in different proportions with group C diluent, the sperm viability and effective survival time under the same preservation time are significantly better than those in other groups when the dilution ratio is 1∶8(P<0.05). [Conclusion] The semen cryopreservation effect is the best when the dilution ratio is 1∶8 when sodium citrate is used as the main buffer adding 15% egg yolk.
