类风湿关节炎患者滑膜组织中GLUT1表达及临床意义
The Expression and Clinical Significance of GLUT1 in Synovial Tissue of Patients with Rheumatoid Arthritis
【摘要】 目的 探究类风湿关节炎(RA)患者滑膜组织中葡萄糖转运蛋白1(GLUT1)表达水平及意义。方法 选取2019年1月至2020年12月本院收治的110例RA患者为RA组,另纳入同期65例半月板损伤者为对照组。实时荧光定量PCR(qRT-PCR)法检测滑膜组织GLUT1 mRNA、低氧诱导因子-1α(HIF-1α)mRNA表达水平;免疫比浊法测定血清类风湿因子(RF)水平;魏氏法测定红细胞沉降率(ESR);对RA患者进行28个关节疾病活动(DAS28)评分;比较缓解组、活动组RA患者滑膜组织GLUT1 mRNA、HIF-1αmRNA、ESR及血清RF水平;Pearson法分析活动期RA患者滑膜组织GLUT1 mRNA、HIF-1αmRNA表达水平与ESR、血清RF水平及DAS28评分的相关性,及GLUT1 mRNA表达水平与HIF-1αmRNA的相关性。结果 RA组患者滑膜组织GLUT1 mRNA、HIF-1αmRNA、ESR、血清RF水平均高于对照组(t=11.464、10.212、15.503、22.999,P<0.05);活动组RA患者滑膜组织GLUT1 mRNA、HIF-1αmRNA、ESR、血清RF水平、DAS28评分高于缓解组(t=3.079、3.526、4.914、9.897、15.822,P<0.05);活动期RA患者滑膜组织GLUT1 mRNA、HIF-1αmRNA表达水平与ESR、血清RF水平及DAS28评分均呈正相关(P<0.05),GLUT1 mRNA表达水平与HIF-1αmRNA呈正相关(r=0.545,P<0.05)。结论 RA患者滑膜组织GLUT1 mRNA表达水平较高,与HIF-1αmRNA、RF、ESR及疾病活动度显著相关,其可能是诊治RA的潜在靶标。
【Abstract】 Objective To explore the expression level and significance of glucose transporter protein 1(GLUT1)in synovial tissue of patients with rheumatoid arthritis(RA).Methods A total of 110 RA patients admitted to our hospital from January, 2019 to December, 2020 were selected as the RA group, while 65 patients with meniscal injury during the same period were included as the control group. Real-time fluorescent quantitative PCR(qRT-PCR)method was used to detect expression levels of GLUT1 mRNA and hypoxia inducible factor-1α(HIF-1α)mRNA in synovial tissue; immunoturbidimetric method was applied to determinate the level of serum rheumatoid factor(RF); westergren method was used to measure erythrocyte sedimentation rate(ESR); 28-joint disease activity(DAS28)scores was performed on the RA patients; levels of GLUT1 mRNA, HIF-1α mRNA, ESR and serum RF in the synovial tissue of RA patients in the remission group and the active group were then compared; Pearson analysis was used to evaluate the correlation between GLUT1 mRNA, HIF-1α mRNA expression levels with ESR, serum RF levels and DAS28 scores in synovial tissues of patients with active RA, and the correlation between GLUT1 mRNA expression level with HIF-1α mRNA.Results Levels of GLUT1 mRNA, HIF-1α mRNA, ESR, and serum RF of synovial tissue in RA group were higher than those in the control group(t=11.464, 10.212, 15.503, 22.999, P<0.05); synovial tissue GLUT1 mRNA, HIF-1α mRNA, ESR, serum RF, and DAS28 scores of RA patients in the active group were higher than those in the remission group(t=3.079, 3.526, 4.914, 9.897, 15.822, P<0.05); expression levels of GLUT1 mRNA and HIF-1α mRNA in synovial tissues of patients with active RA were positively correlated with ESR, serum RF level and DAS28 scores(P<0.05), the expression level of GLUT1 mRNA was positively correlated with HIF-1α mRNA(r=0.545, P<0.05).Conclusion The expression level of GLUT1 mRNA in synovial tissues of RA patients is high, and it is significantly related to HIF-1α mRNA, RF, ESR and disease activity, which could serve as a potential target for the diagnosis and treatment of RA.
