RNF187在脑胶质瘤中的表达及其与预后生存的关系
Study on the expression of RNF187 and its association with prognosis and survival of glioma patients
【摘要】 目的探索环指蛋白187(RNF187)在脑胶质瘤中的表达及其与预后生存的关系。方法选取2015年1月至2018年10月于本院术后病理为脑胶质瘤患者49例,按胶质瘤病理分级分为低度恶性神经胶质瘤组(LM组,18例)和高度恶性神经胶质瘤组(HM组,31例),收集两组受检者的肿瘤组织并检测RNF187和肿瘤标志物Notch1的mRNA和蛋白,并随访患者术后2年生存情况;随后分析RNF187与Notch1的关系,以及与胶质瘤分级和临床预后的关系;最后通过U251细胞转染沉默RNF187,对比分析RNF187沉默对肿瘤细胞生长增殖能力、侵袭能力的影响,以及对Notch1表达的影响。结果 RNF187 mRNA与蛋白水平在HM组高于LM组,与肿瘤分级呈正相关,与临床预后呈负相关,与Notch1表达正相关,差异均有统计学意义(P<0.05);RNF187高表达意味着更低预后生存期;沉默RNF187后U251细胞增殖和侵袭能力较对照组明显下降,Notch1表达明显减少,差异均有统计学意义(P<0.05)。结论 RNF187与脑胶质瘤恶性度正相关,沉默RNF187后可以通过降低Notch1表达减轻胶质瘤细胞的恶性度,为脑胶质瘤的临床诊断和治疗提供新的依据。
【Abstract】 Objective To investigate the expression of ring finger protein 187(RNF187) and its relationship with prognosis and survival of glioma patients.Methods From January 2015 to October 2018,49 patients with postoperative pathological brain glioma were selected and divided into low-grade malignant glioma group(LM group, 18 cases) and high-grade malignant glioma group(HM group, 31 cases) according to the pathological grade of glioma.The tumor tissues of the two groups were collected and the expressions of RNF187 and Notch1 were detected.Then, the relationship between RNF187 and Notch1,as well as the relationship between RNF187 and glioma grade and clinical prognosis respectively, were analyzed Finally, the effects of RNF187 silencing on tumor cell growth, proliferation, invasion and Notch1 expression of U251 cells were detected.Results The levels of RNF187 mRNA and protein in HM group were higher than those in LM group, which were positively correlated with tumor grade, negatively correlated with clinical prognosis, and positively correlated with Notch1 expression, respectively(P<0.05).Moreover, the high expression of RNF187 meant lower survival time.Finally, after silencing RNF187,the proliferation and invasion ability of U251 cells were significantly decreased, accompanied with reduced expression level of Notch1(P<0.05).Conclusion Silencing RNF187 can reduce the malignant degree of glioma cells by reducing the expression of Notch1,which provides a new basis for clinical diagnosis and treatment of glioma.
