芒柄花黄素通过circ0020123/miR-145对胰腺癌细胞增殖迁移侵袭的影响

【摘要】 目的:探讨芒柄花黄素对胰腺癌细胞的影响及分子机制。方法:将胰腺癌细胞SW1990分为对照组、芒柄花黄素低、中、高剂量组(10,30,100μmol·L-1)、si-circ0020123组、si-NC组、芒柄花黄素(100μmol·L-1)+pcDNA-circ0020123组、芒柄花黄素(100μmol·L-1)+pcDNA组;蛋白质印迹(Western blot)法检测蛋白表达;四甲基偶氮唑盐比色法(MTT)检测细胞增殖抑制率;克隆形成实验检测集落形成数;划痕实验检测细胞划痕愈合率;Transwell检测侵袭细胞数;实时荧光定量PCR(RT-qPCR)检测circ0020123和miR-145的表达水平;双荧光素酶报告实验检测circ0020123和miR-145的靶向关系。结果:不同剂量芒柄花黄素处理后,胰腺癌SW1990细胞的增殖抑制率、E-钙黏蛋白(E-cadherin)及miR-145表达水平升高,集落形成数和侵袭细胞数减少,细胞划痕愈合率、N-钙黏蛋白(N-cadherin)及circ0020123表达水平降低(P<0.05)。干扰circ0020123表达后,细胞增殖抑制率、E-cadherin及miR-145表达水平升高,集落形成数和侵袭细胞数减少,细胞划痕愈合率、N-cadherin及circ0020123表达水平降低(P<0.05)。circ0020123过表达可减弱芒柄花黄素对SW1990增殖迁移侵袭的抑制作用。circ0020123靶向调控miR-145。结论:芒柄花黄素通过调控circ0020123/miR-145抑制胰腺癌细胞的增殖与转移。
【Abstract】 Objective: To explore the effects and its molecular mechanism of formononetin on pancreatic cancer cells. Methods: Pancreatic cancer cells SW1990 were divided into control group, formononetin low, medium and high dose groups(10,30,100 μmol·L-1), si-circ0020123 group, si-NC group, formononetin(100 μmol·L-1)+pcDNA-circ0020123 group and formononetin(100 μmol·L-1)+pcDNA group; Western blot was used to detect protein expression; tetramethylazolium salt colorimetric method(MTT) was used to detect cell proliferation inhibition rate; clone formation experiment was used to detect colony formation number; scratch experiment was used to detect cell scratch healing rate; Transwell was employed to detect the number of invaded cells; real-time fluorescent quantitative PCR(RT-qPCR) was used to detect the expression levels of circ0020123 and miR-145; dual luciferase reporter experiment was used to detect the targeting relationship between circ0020123 and miR-145.Results: After treatment with different doses of formononetin, the proliferation inhibition rate, E-cadherin and miR-145 expression levels of pancreatic cancer SW1990 cells increased, and the number of colonies formed, number of invasive cells, cell scratch healing rate, N-cadherin and circ0020123 expression levels decreased(P<0.05). After interference with the expression of circ0020123, the proliferation inhibition rate, E-cadherin and miR-145 expression levels increased and the number of colonies formed, number of invasive cells, cell scratch healing rate, N-cadherin and circ0020123 expression levels decreased(P<0.05). Overexpression of circ0020123 could attenuate the inhibitory effects of formononetin on the proliferation, migration and invasion of SW1990. circ0020123 targeted and regulated miR-145. Conclusion: Formononetin inhibits the proliferation and metastasis of pancreatic cancer cells by regulating circ0020123/miR-145.
