电针对创伤性脑损伤大鼠GLT-1和细胞凋亡的影响

【摘要】 目的观察电针对创伤性脑损伤(TBI)大鼠脑组织中谷氨酸转运蛋白-1(GLT-1)的表达和细胞凋亡的影响,探讨电针对创伤性脑损伤的脑保护作用。方法将40只SD大鼠随机分成假手术组、TBI组、手针组、电针组4组,每组10只。假手术组和TBI组不予以干预,手针组和电针组在造模后立即干预,共7 d,所有大鼠分别在干预第2天、第4天、第7天进行mNSS评估神经功能。第7天干预结束后采用HE染色观察炎性细胞浸润情况、TUNEL检测细胞凋亡情况,实时荧光定量PCR检测脑组织GLT-1 mRNA的表达情况。结果 1)mNSS:在干预第2天,TBI组、手针组、电针组mNSS评分均高于假手术组,且TBI组与假手术组相比有统计学差异(P <0.01);在第4天和第7天,手针组和电针组评分虽都降低,但电针组比TBI组减少更为显著(P <0.01)。2)HE染色和TUNEL:与假手术组相比,TBI组和手针组炎性细胞数和凋亡细胞数均减少且有统计学意义(P <0.01或P <0.05);与TBI组相比,电针组细胞数量显著降低(P <0.05)。3)实时荧光定量PCR:与假手术组相比,TBI组GLT-1 mRNA含量明显降低(P <0.05),电针组含量明显升高(P<0.01);与TBI组比较,电针组和手针组GLT-1mRNA表达升高(P <0.01或P <0.05);电针组的GLT-1 m RNA的表达量显著高于手针组(P <0.01)。结论针刺可提高TBI大鼠脑内GLT-1 m RNA的表达,抑制谷氨酸兴奋性毒性,减轻神经炎症,降低细胞凋亡和改善神经功能评分,从而减轻脑损伤达到保护脑的作用,且电针的干预效果优于手针。

【Abstract】 Objective:To observe the effects of electroacupuncture on the expression of glutamate transporter-1(GLT-1)and apoptosis in Traumatic brain injury(TBI)rats,and to explore the protective effect of electroacupuncture on brain. Methods:A total of 40 SD rats were randomly divided into 4 groups:sham group,TBI group,manual acupuncture(AC)group and electroacupuncture(EA)group,10 rats in each group. The sham group and the TBI group did not receive intervention,while the AC group and the EA group received intervention immediately after modeling for a total of 7 days. m NSS were performed on the 2 nd,4 th and 7 th day with all rats. On the 7 th day after treatment,HE staining was used to observe the inflammatory cells infiltration,apoptosis was observed by TUNEL,and GLT-1 mRNA expression was observed by real-time PCR. Results:1)mNSS:On the 2 nd day of intervention,mNSS scores in the TBI group,AC group and EA group were all higher than those in the sham group,and there was a statistical difference between the TBI group and the sham group(P < 0.01). On the 4 th and 7 th day,scores in both the AC group and EA group decreased,but the reduction was more significant in EA group than in the TBI group(P < 0.01). 2)HE staining and TUNEL:Compared with sham group,the number of inflammatory cells and apoptotic cells were decreased in TBI group and AC group,with statistical significance(P < 0.01 or P <0.05). Compared with TBI group,the number of cells in EA group was significantly decreased(P < 0.05). 3)Realtime quantitative PCR:Compared with Sham group,the content of GLT-1 mRNA in TBI group was significantly decreased(P < 0.05),and the content of GLT-1 mRNA in EA group was significantly increased(P < 0.01). Compared with TBI group,the expression of GLT-1 m RNA in EA group and AC group was increased(P < 0.01 or P <0.05). The expression of GLT-1 mRNA in EA group was significantly higher than that in AC group(P < 0.01).Conclusion:Acupuncture can increase the expression of GLT-1,inhibit the excitatory toxicity of glutamate,reduce neuroinflammation,reduce cell apoptosis and improve the neurological function score,so as to reduce the brain injury and protect the brain,and the intervention effect of electroacupuncture is better than manual acupuncture.
