清肝降浊方基于TLR4/NF-?B信号通路治疗非酒精性脂肪肝病作用机制研究
An investigation of the mechanism of Qinggan Jiangzhuo Particles in the treatment of non-alcoholic fatty liver disease based on the TLR4/NF-κB signaling pathway
【摘要】 目的通过试验探讨清肝降浊方基于TLR4/NF-κB信号通路治疗非酒精性脂肪肝病(NAFLD)的作用机制。方法采用高脂饮食饲喂的方法,诱导建立非酒精性脂肪肝病大鼠动物模型。预防性给药12周后,检测血脂(TC、TG)、肝脂(TC、TG)及肝功能(AST、ALT)指标,肝脏病理组织学检查,Western-blot检测信号通路相关蛋白TLR4、NF-κB P65、MyD88表达水平。结果清肝降浊方3个给药组动物血清AST、ALT活性明显低于模型组、大鼠肝组织中TLR4表达量显著低于模型对照组(P<0.01或P<0.05);高、中给药组大鼠肝组织匀浆及血清中TG、TC浓度明显低于模型组、大鼠肝组织中NF-κB P65、MyD88表达量显著低于模型对照组(P<0.01或P<0.05);清肝降浊方3个给药剂量组都可不同程度地减轻肝脏病理损伤。结论清肝降浊方具有保肝降酶、降低血脂、肝脂含量,减轻肝脏病理损伤作用,可能是通过调节TLR4/NF-κB信号通路中TLR4、NF-κB P65、MyD88蛋白表达,起到防治非酒精性脂肪肝病作用。
【Abstract】 Objective To investigate the mechanism of Qinggan Jiangzhuo Particles in the treatment of non-alcoholic fatty liver disease (NAFLD) based on the TLR4/NF-κB signaling pathway.Methods A non-alcoholic fatty liver disease rat model was induced by feeding them high fat diet.After 12 weeks of preventive administration,the indexes of serum lipid (TC,TG),liver lipid (TC,TG) and liver function (AST,ALT) were detected,the liver histopathology was examined,and the expression level of signaling pathway of related proteins TLR4,NF-κB P65 and MyD88 was detected by Western-blot.Results The activities of serum AST and ALT in the three administration groups were significantly lower than those of the model group,and the expression of TLR4 in rat liver tissues was significantly lower than that of the control group (P<0.01 or P<0.05).The TG and TC concentration in liver tissue homogenate and serum of rats in the high and medium administration groups was significantly lower than that in the model group,and the expression of NF-κB P65 and MyD88 in liver tissues was significantly lower than that in the control group (P<0.01 or P<0.05).The pathological damage of the liver in the three administration groups was alleviated to varying degrees.Conclusion Qinggan Jiangzhuo Particles can protect the liver and lower enzymes,reduce the content of blood lipid and liver lipid,and alleviate liver pathological damage,possibly by regulating the expression of TLR4,NF-κB P65 and MyD88 proteins in TLR4/NF-κB signaling pathway to prevent and treat the disease.
