【摘要】 目的:基于"病证结合"原则,复合运用慢性不可预知温和应激(chronic unpredictable mild stress, CUMS)和大黄灌胃构建抑郁症肝郁脾虚证的小鼠模型,探索模型小鼠血浆γ氨基丁酸(GABA)代谢的变化,为抑郁症肝郁脾虚证的研究提供参考。方法:将29只成年雄性小鼠随机分为对照组14 d和实验组15 d,对实验组连续14 d施行CUMS,自第15天开始除每天CUMS外增加大黄灌胃(15%,0.5ml/d)直至28d造模结束,对照组不做任何处理。观察比较小鼠的体质量增加、行为学改变及大便性状的改变和血浆GABA代谢相关特征。结果:2组小鼠在体质量增加、行为学改变、粪便性状和血浆GABA代谢的改变方面差异显著。结论:CUMS合并大黄灌胃构建小鼠模型符合抑郁症肝郁脾虚证表现,可应用于抑郁症肝郁脾虚证的基础与临床研究。更多还原

【Abstract】 Objective: To construct a mouse model of depression of liver depression and spleen deficiency syndrome by Chronic Unpredictable mild stress CUMS and Rhubarb gavage based on the principle of combining disease with syndrome, and the changes of plasma GABA metabolism in the model mice were explored to provide reference for the study of depression with liver depression and spleen deficiency syndrome. Methods:29 adult male mice were randomly divided into control group(n=14) and experimental group(n=15). CUMS was administered to experimental group for 14 consecutive days, and Rhubarb gavage(15%, 5 ml/d) was increased in addition to daily CUMS from day 15 until the end of modeling on day 28. The control group did not receive any intervention. And the weight gain, behavior change and fecal character change of two groups were compared. Results: There were significant differences between the two groups in weight gain, behavioral changes,fecal traits and concentration of plasma GABA/GAD65/GAD67. Conclusion: The mouse model of depression of liver depression and spleen deficiency syndromes constructed by CUMS combined with Rhubarb gavage conforms to the manifestation of depression of liver-depression and spleen-deficiency syndromes, and has clinical and basic research value.更多还原
