miR-34a及其下游因子Bcl-2、TGF-β1在胆管癌发展中的作用
Role of miR-34a, Bcl-2 and TGF-β1 in the Development of Cholangiocarcinoma
【摘要】 目的:研究miR-34a调控网络在胆管癌发展中的作用及对化疗的影响。方法:收集2014年6月-2018年6月86例本院收治的胆管癌患者(试验组)及86例健康人(对照组)的资料,并取其血清利用实时定量聚合酶链式反应(qRT-PCR)测试miR-34a表达水平,同时检测调控因子Bcl-2、TGF-β1的表达量。结果:试验组miR-34a、Bcl-2及TGF-β1表达量均明显高于对照组,差异均有统计学意义(P<0.001);不同miR-34a表达程度胆管癌患者性别、年龄、分化程度比较,差异均无统计学意义(P>0.05);miR-34a高表达患者TNM分期、Bcl-2、TGF-β1与miR-34a低表达患者比较,差异均有统计学意义(P<0.001)。试验组化疗后miR-34a、TGF-β1及Bcl-2表达量均明显低于化疗前,差异均有统计学意义(P<0.001),胆管癌患者血清miR-34a与Bcl-2、TGF-β1均呈正相关(r=0.872、0.876,P=0.041、0.003)。结论:胆管癌患者miR-34a表达量明显高于健康人,可能与调控下游Bcl-2、TGF-β1的表达量有关,此外miR-34a、Bcl-2及TGF-β1随患者的治疗活动变化产生相应改变,可见三者可作为胆管癌诊疗的潜在位点。
【Abstract】 Objective:To study the role of miR-34a regulatory network in the development of cholangiocarcinoma and its influence on chemotherapy.Method:The data of 86 patients with cholangiocarcinoma (the experimental group) and 86 healthy people (the control group) from June 2014 to June 2018 were collected.The expression of miR-34a was detected by real-time quantitative polymerase chain reaction (qRT-PCR),and the expressions of Bcl-2 and TGF-β1 were detected.Result:The expressions of miR-34a,Bcl-2 and TGF-β1 in the experimental group were significantly higher than those in the control group (P<0.001).There were no significant differences in gender,age and differentiation degree of cholangiocarcinoma patients with different miR-34a expression levels (P>0.05).There were significant differences in TNM stage,Bcl-2 and TGF-β1 between patients with high expression of miR-34a and patients with low expression of miR-34a (P<0.001).The expression of miR-34a,Bcl-2 and TGF-β1 of the experimental group after chemotherapy were significantly lower than those of before chemotherapy (P<0.001).And there were positive correlations between serum miR-34a and Bcl-2,TGF-β1 (r=0.872,0.876;P=0.041,0.003).Conclusion:The expression of miR-34a in patients with cholangiocarcinoma is significantly higher than that in healthy people,which may be related to the regulation of the expression of downstream Bcl-2 and TGF-β1.In addition,miR-34a,Bcl-2 and TGF-β1 have corresponding changes with the changes of patients’ treatment activities.It can be seen that the three can be used as potential points for diagnosis and treatment of cholangiocarcinoma.
