HPLC同步测定秃疮花中5种生物碱的含量

【摘要】 目的:建立HPLC(1525-2707TCM-2489色谱系统)同时测定秃疮花中5种生物碱含量方法。方法:采用工业乙醇超声辅助法提取秃疮花全草干粉制得乙醇提取物,再用等量石油醚、氯仿、正丁醇依次萃取,然后采用柱层析法逐步分离得到紫堇丁碱、异紫堇丁碱、黄连碱、木兰花碱、小檗碱,采用HPLC测定其含量。采用HPLC、柱型C18、甲醇(A)-水(B)为流动相,梯度洗脱(0~5 min, 10%A;5~35 min, 10%A→100%A;45~48 min, 100%A→10%A,48~50 min, 10%A),上样量1 mL,进样体积10μL,每个样品运行检测时间50 min;柱温30℃,检测波长260 nm。结果:紫堇丁碱的含量是7.90 mg/g,异紫堇丁碱的含量是13.30 mg/g,黄连碱的含量是1.36 mg/g,木兰花碱的含量是1.15 mg/g,小檗碱的含量是0.76 mg/g。结论:紫堇丁碱和异紫堇丁碱是秃疮花酸化氯仿相的主要生物碱类活性成分,黄连碱和木兰花碱是秃疮花正丁醇相中的主要生物碱类活性成分。该试验为定量测定药用植物中单体化合物的含量奠定了实验基础,可为同源植物系统研究提供理论参考。

【Abstract】 Objective: To establish an HPLC method for the simultaneous determination of 5 alkaloids in Dicranostigma lptopodum(Maxim.) Fedde. Method: The ethanol extract was prepared from the whole herb powder of Dicranostigma lptopodum(Maxim.) Fedde by ultrasonic assisted extraction with industrial ethanol. After extraction with petroleum benzine, chloroform and n-butanol,(+)Corydrine,(+)Isocorydrine, Coptisine, Magnoflorine and Berberine were obtained by column chromatography, whose contents were determined by HPLC. Chromatographic condition was that HPLC(E2695 Waters, 1525-2707 TCM-2489), C18 column(250 mm×4.6 mm, 5 μm), methanol(A)-water(B) mobile phase, gradient elution(0~5 min, 10% A; 5~35 min, 10% A→100% A; 45~48 min, 100% A→10% A; 48~50 min, 10% A), the sample loading volume 1 mL, the injection volume 10 μL, the detection time of each sample 50 min, the column temperature 30 ℃, the detection wavelength 260 nm. Results: The content of Corydrine was 7.90 mg/g; Isocorydrine, 13.30 mg/g; Coptisine, 1.36 mg/g; Magnoflorine, 1.15 mg/g; Berberine, 0.76 mg/g. Conclusions: Corydrine and isocorydrine are the main active components in the acidified chloroform phase, while Coptisine and Magnoflorine are the main alkaloids in the n-butanol phase of Dicranostigma lptopodum(Maxim.) Fedde. This experiment lays an experimental foundation for the quantitative determination of monomer compounds in medicinal plants, and can provide theoretical reference for the systematic study of homologous plants.
