miR-133a水平降低为非小细胞肺癌的不良预后因子

【摘要】 目的探讨非小细胞肺癌(NSCLC)患者的MicroRNA-133a(miR-133a)表达水平,及其与患者临床病理特征和预后的关系。方法收集南通市第一人民医院2017年1月-2019年10月的NSCLC患者82例为研究对象。使用定量实时PCR分析方法检测并分析miR-133a在研究对象肺癌组织和邻近正常肺组织中的表达。使用ROC曲线分析miR-133a对NSCLC及病理特点的预测价值。使用生存曲线分析miR-133a对NSCLC患者生存时间的影响。结果 NSCLC组织的miR-133a表达降低(NSCLC:0.25±0.12,邻近正常肺组织:0.68±0.14;P<0.001)。ROC曲线分析表明miR-133a对NSCLC、患者进展至癌症晚期、患者发生淋巴转移有一定的预测价值,曲线下面积分别为0.7474(P<0.001)、0.6613(P<0.001)、0.6532(P<0.001)。生存曲线表明高miR-133a表达组的生存时间高于低水平表达组(19.48±5.48月vs.16.74±4.38月,P=0.058)。结论 miR-133a可作为NSCLC的标志物,其表达降低提示NSCLC患者预后不良。

【Abstract】 Objective To investigate the expression level of MicroRNA-133 a(miR-133 a)in patients with non-small cell lung cancer(NSCLC)and its relationship with clinicopathological characteristics and prognosis of patients.Methods A total of 82 patients with NSCLC from January 2017 to October 2019 in our hospital were collected as the research subjects.Quantitative real-time PCR analysis was used to detect and compare the expression of miR-133 ain lung cancer tissues and adjacent normal lung tissues.The ROC curve was used to analyze the predictive value of miR-133 afor NSCLC and pathological characteristics.The survival curve was used to analyze the effect of miR-133 aon the survival time of NSCLC patients.Results The expression of miR-133 ain NSCLC tissue was decreased(NSCLC:0.25±0.12,adjacent normal lung tissue:0.68±0.14;P<0.001).ROC curve analysis showed that miR-133 ahad certain predictive value for NSCLC,progression to advanced stage,and lymphatic metastasis.The areas under the curve were 0.747 4(P<0.001),0.661 3(P<0.001),and 0.653 2(P<0.001),respectively.The survival curve showed that the survival time of the high miR-133 aexpression group was higher than that of the low-level expression group(19.48±5.48 months vs.16.74±4.38 months,P=0.058).Conclusion miR-133 acan be used as a marker for NSCLC,and its reduced expression suggests that NSCLC patient has a poor prognosis.miR-133 amay be a promising predictive biomarker and possible therapeutic target for non-small cell lung cancer.
