不同感染阶段奶牛粪便副结核分枝杆菌的排逸状况分析

【摘要】 为探索奶牛感染副结核分枝杆菌的粪便排菌情况,本试验提取血清抗体阳性奶牛的粪便DNA,通过Real-time PCR试剂盒进行核酸检测,综合分析不同感染阶段奶牛粪便中副结核分枝杆菌的排逸状况。结果显示,血清抗体阳性牛可通过粪便排放副结核分支杆菌,粪便检出率为5.82%(11/189),其中亚临床期牛的粪便检出率为2.98%(5/168),而临床期牛的粪便检出率为28.57%(6/21),且临床期粪便排放的副结核分支杆菌数量为亚临床期的32倍。鉴于感染副结核的奶牛会通过粪便排放致病菌到环境中,建议牛场标记感染牛,并在出现腹泻症状时淘汰。

【Abstract】 In order to investigate the fecal excretion status of Mycobacterium avium subsp. paratuberculosis(MAP)-antibody seropositive dairy cattle, this study extracted the fecal DNA, and nucleic acid detection was carried out by PCR kit to comprehensively analyze the excretion status of MAP in the faeces of dairy cows at different infection stages. The results showed that 5.82%(11/189) fecal samples from all the infected cattle, 2.98%(5/168) fecal samples from the subclinical infected cattle, and 28.57%(6/21) fecal samples from the clinical infected cattle contained detectable MAP DNA, with the gene copy number from clinical infected cattle being 32 times higher than that from subclinical infected cattle. These results provide the evidence that infected cattle can excrete MAP into environment, so dairy cattle farms should mark the infected cattle and cull them when they begin diarrhea.
