1例犬乳腺癌的病理组织学诊断
Histopathological Diagnosis of a Canine Breast Cancer
【摘要】 为了鉴定1例犬乳腺肿物性质及病理变化,本试验利用苏木素-伊红(H.E.)染色及免疫组化等方法对1例犬乳腺肿瘤进行病理组织学检查,分析肿瘤组织细胞的病理变化。通过细胞分离培养,确定肿瘤细胞的生长特性;并用免疫荧光法检测肿瘤细胞标记物,判断该病例的预后情况。H.E.染色结果显示,该病例乳腺细胞和乳腺导管发生癌变。免疫组化结果显示,肿瘤相关蛋白P63、CK5/6及α-SMA表达阳性,雌激素(ER)、孕激素(PR)、人表皮生长因子-2(HER-2)的蛋白表达均为阴性,表明该病例为三阴性犬乳腺癌。纯化的癌细胞呈上皮细胞特征,细胞倍增时间为26.35 h。该细胞的免疫荧光检测结果显示,SOX-2及Ki-67高表达,进一步佐证其为乳腺癌,并且预后不良。免疫组化试验结果与组织病理学判定结果一致,表明此犬乳腺肿物为三阴性乳腺癌。
【Abstract】 In order to identify the nature and pathological changes of a canine breast tumor, this experiment used hematoxylin-eosin(H.E.) staining and immunohistochemistry to conduct histopathological examination of a canine breast tumor to analyze the pathological changes of tumor tissue cells. Determined the growth characteristics of tumor cells through cell separation and culture, and used immunofluorescence to detect tumor cell markers to determine the prognosis of the case. The H.E. staining results showed that the breast cells and mammary ducts were cancerous. The immunohistochemistry results showed that P63, CK5/6 and α-SMA were positive; the expressions of estrogen(ER), progesterone(PR) and human epidermal growth factor-2(HER-2) were negative, indicating that the breast cancer was a triple negative canine breast cancer. Purified cells were characterized by epithelial cells and its doubling time was 26.35 h. The results of immunofluorescence showed that SOX-2 and Ki-67 were highly expressed, which further confirmed that it was breast cancer with poor prognosis. The results of immunohistochemistry were consistent with histopathological results. In conclusion, this canine mammary tumor was a triple negative breast cancer.
