齐整小核菌发酵产胞外多糖的培养基优化研究

【摘要】 为提高齐整小核菌(Sclerotium rolfsii)发酵产胞外多糖能力,该研究对培养基不同碳源、不同氮源和无机盐进行优化,采用单因素试验、最陡爬坡试验和响应面(Box-Behnken)设计试验等方法优选培养基配方。结果表明,最优培养基配方为葡萄糖136 g/L、胰蛋白胨7 g/L、硝酸钠4 g/L、氯化钾0.5 g/L、磷酸氢二钾1.3 g/L、硫酸镁0.4 g/L和硫酸亚铁0.1 g/L。在此优化条件下,多糖产量最高,为(23.76±1.05)g/L,相比原始培养基提高54.42%。

【Abstract】 To improve the exopolysaccharides-producing ability of Sclerotium rolfsii, different carbon sources, nitrogen sources and inorganic salts of the medium were optimized. The medium formula was optimized by single falfor experiments, climbing experiments and Box-Behnken design experiments.The results showed that the optimal medium was obtained as glucose 136 g/L, tryptone 7 g/L, Na NO34 g/L, KCl 0.5 g/L, K2 HPO41.3 g/L, Mg SO40.4 g/L and Fe SO40.1 g/L. Under the optimal conditions, the maximum polysaccharide yield was(23.76±1.05) g/L, which was 54.42% higher than that of the original medium.
