不同分子量的枸杞多糖组分对树突状细胞成熟的影响
Effects of Lycium barbarum polysaccharides with different molecular weights on maturation of dendritic cells
【摘要】 目的:本研究旨在筛选诱导树突状细胞(DCs)成熟活性最强的枸杞多糖(LBPs)组分。方法:体外诱导培养未度,CBA法检测培养上清中细胞因子(IL-6、IL-10、MCP-1、TNF-α和IFN-γ)的分泌情况。流式细胞术检测LBP3刺激后DCs表著。LBP3可明显上调DCs表面分子CD11c、MHCⅡ、CD80和CD86表达,与对照组相比,差异具有统计学差异(P<0.01)。结论:中等分子量的LBP3是LBPs中诱导DCs成熟的主要活性成分。
【Abstract】 Objective:This study was aimed to screen the most active Lycium barbarum polysaccharides(LBPs)components on inducing maturation of dendritic cells(DCs).Methods:Immature DCs were generated from bone marrow cells in vitro,and treated with different doses of LBPs fractions(LBP2,LBP3,LBP4,LBP5). Concentration of nitric oxide(NO)and cytokines(IL-6,IL-10,MCP-1,TNF-α and IFN-γ)in the culture supernatant were detected by Griess reagent or CBA. Flow cytometry was used to detect the phenotype of DCs stimulated by LBP3.Results:LBP3,LBP4 and LBP5 could stimulate DCs to product NO,IL-6,IL-10,MCP-1,TNF-α and IFN-γ,and LBP3 was the most active ingredient in promoting the production of NO and the cytokines. LBP3 upregulated expression of CD11 c,MHCⅡ,CD80 and CD86,which were statistically different compared with control group(P<0.01).Conclusion:LBP3 with a medium molecular weight was the most active ingredient in inducing the maturation of DCs.
