白藜芦醇对烟草烟雾提取物诱导的骨骼肌细胞衰老的保护作用及机制

【摘要】 目的研究白藜芦醇(Res)对烟草烟雾提取物(CSE)所致骨骼肌细胞衰老的保护作用及其机制。方法将C2C12成肌细胞培养分化为骨骼肌细胞,用CSE诱导细胞衰老,分别用Res、N-乙酰半胱氨酸(NAC)和过氧化氢(H2O2)干预,β-半乳糖苷酶染色法检测衰老细胞阳性率,实时荧光定量聚合酶链反应(PCR)和Western印迹法分别检测抗衰老分子组蛋白去乙酰化酶(HDAC)2 mRNA和蛋白水平,观察骨骼肌细胞中氧化应激产物丙二醛(MDA)含量、谷胱甘肽过氧化物酶(GSH-Px)活性、活性氧(ROS)水平和超氧化物歧化酶(SOD)活性的变化。结果与空白对照组比较,CSE和H2O2可促进骨骼肌细胞的衰老,同时伴有骨骼肌细胞中ROS水平和MDA含量升高,GSH-Px、SOD的活性减弱,HDAC2 mRNA及蛋白的水平降低,差异有统计学意义(P<0.05)。和CSE组相比较,Res干预后,骨骼肌细胞的衰老下调,同时MDA含量降低,ROS水平下降,而GSH-Px和SOD的活性增强,HDAC2的mRNA及蛋白水平提高,差异有统计学意义(P<0.05)。与NAC比较,Res抗衰老作用更明显,在增加SOD活性和提高HDAC2 mRNA水平方面作用更强(P<0.05)。结论 Res通过减少CSE诱导的骨骼肌细胞氧化应激和提高抗衰老分子HDAC2的表达来延缓骨骼肌细胞衰老,其作用强于NAC。

【Abstract】 Objective To study the protective effect and the mechanism of resveratrol on senescence of skeletal muscle cells induced by cigarette smoke extract(CSE).Methods C2 C12 myoblasts were induced and differentiated into skeletal muscle cells. The skeletal muscle cells were induced cell senescence by cigarette smoke extract and were incubated with resveratrol(Res), N-acetyl cysteine(NAC) and hydrogen peroxide(H2O2) respectively. The β-galactosidase staining was used to detect the positive rate of aging cells. The mRNA and protein levels of anti-aging molecule histone deacetylase(HDAC)2 were detected by real-time fluorescence quantitative PCR and Western blot respectively. Indicators of oxidative stress, the content of malondialdehyde(MDA), and the activities of glutathione peroxidase(GSH-Px), reactive oxygen species(ROS) and superoxide dismutase(SOD) were observed.Results Compared with the control group, the senescence were increased in skeletal muscle cells after CSE or H2O2 treated, the content of MDA and the activity of ROS were increased, the activity of GSH-Px and SOD were decreased, and the mRNA and protein level of HDAC2 were decreased(P<0.05). Compared with CSE, after intervened with Res, the senescence of skeletal muscle cells was decreased, the content of MDA was decreased, the activity of ROS was weakened, the activities of SOD and GSH-Px were enhanced instead, and the mRNA and protein level of HDAC2 were increased(P<0.05). Compared with NAC, Res had a more obvious anti-aging effect and a stronger role in increasing SOD activity and the level of HDAC2 mRNA(P<0.05).Conclusions Res could delay the senescence of skeletal muscle cells by reducing the oxidative stress of skeletal muscle cells induced by CSE and increasing the expression of anti-aging molecule HDAC2. Its effect is stronger than that of NAC.
