香水白掌的组培快繁技术研究
Study on Tissue Culture and Rapid Propagation of Spathiphyllum patinii
【摘要】 以香水白掌(Spathiphyllum patinii)幼嫩茎段为试材,进行组培快繁技术的探讨。结果表明,最佳外植体消毒方法为流水冲洗2 h,0.05%NaClO处理20 min,转入超净工作台后75%酒精处理20 min,0.1%HgCl2处理15 min。诱导不定芽的最适宜条件为MS+3 mg·L-1 6-BA+0.05 mg·L-1 NAA培养基,光照2 000 lx,温度25~28℃。获得最大不定芽增殖系数的培养条件为MS+0.2 mg·L-1 Kt+2 mg·L-1 6-BA+0.2 mg·L-1 NAA培养基,温度28℃。获得最大不定芽苗高的增殖培养条件为MS+1 mg·L-1 6-BA+0.3 mg·L-1 NAA培养基,温度28℃。将苗高2~4 cm的幼苗转入1/2 MS+0.2 mg·L-1 NAA+0.2 mg·L-1 IBA培养基中,生根率可达100%。
【Abstract】 In this study, the technique of tissue culture and rapid propagation of Spathiphyllum patinii was studied by using young stem segment as experimental material. The results showed that the best disinfection methods were as follows water washing for 2 h, 0.05% NaClO treatment for 20 min, 75% alcohol treatment for 20 min and 0.1% HgCl2 treatment for 15 min after being transferred to the ultra-clean worktable. The optimum conditions for adventitious bud induction were MS medium+3 mg·L-1 6-BA+0.05 mg·L-1 NAA, illumination 2 000 lx, temperature 25—28 ℃. The maximum adventitious bud proliferation coefficient was obtained under the conditions of MS+0.2 mg·L-1 KT+2 mg·L-1 6-BA+0.2 mg·L-1 NAA at 28 ℃. The maximum adventitious shoot height was obtained on MS medium+1 mg·L-1 6-BA+0.3 mg·L-1 NAA at 28 ℃. The rooting rate of 2—4 cm seedlings was 100% when transplanted into 1/2 MS medium with 0.2 mg·L-1 NAA and 0.2 mg·L-1 IBA.
