河北北部地区PCV4流行株的遗传变异分析

【摘要】 为了解河北省猪圆环病毒4型(PCV4)分子流行病学特点和遗传进化规律,本研究对2016年以来收集自河北北部地区395份临床猪血清、肺脏、脾脏等样品,采用PCR技术进行PCV4检测,并对9株PCV4的全基因组进行了扩增、克隆和测序,利用Megalign等软件分析其与参考株全基因组的遗传进化关系。结果显示,395份样品中,86份检测为PCV4阳性,阳性率21.8%,9株分离株与当前流行的PCV4参考株序列同源性高达98.1%~99.8%,与PCV1/2/3参考株相应序列同源性为42.0%~52.2%,与狐圆环病毒(Fox CV)、蝙蝠圆环病毒(Bat CV)和水貂圆环病毒(Mink CV)全基因组序列同源性分别为52.6%~52.9%,39.3%~39.5%和67.9%~68.7%。从遗传进化关系上,9株PCV4株与Mink CV属于同一分支,亲缘关系最近。本研究首次对河北北部地区进行了PCV4分子流行病学调查,证实河北地区存在PCV4流行,丰富了河北乃至我国PCV4的流行病学数据,为PCV4感染的防控提供了重要的参考价值。

【Abstract】 In order to investigate the molecular epidemiology characteristics and genetic variation of porcine circovirus type 4(PCV4) in Hebei province, 395 clinical porcine samples, including serum, lung, spleen, were collected in northern Hebei province since 2016. PCV4 genome was detected by PCR and the whole genome of 9 strains was amplified, cloned and sequenced, then the genetic evolution relationship between the 9 strains and reference strains was analyzed by Megalign software etc. The results showed that 86 out of 395 samples were positive for PCV4, with a positive rate of 21.8%. The nine isolated strains shared a nucleotide similarity of 98.1%-99.8% among the current epidemic PCV4 reference strains. Furthermore, the corresponding sequence homology with PCV1/2/3, Fox CV, Bat CV and Mink CV were 42.0%-52.2%, 52.6%-52.9%, 39.3%-39.5%, 67.9%-68.7%. In terms of genetic evolution, 9 PCV4 isolated strains and Mink CV belong to the same branch, indicating that they are closely related. In this study, molecular epidemiological investigation of PCV4 was conducted in northern Hebei for the first time,which confirmed the prevalence of PCV4 in Hebei. The study not only enriches the epidemiological data of PCV4 in Hebei and even in China, but also provides an important reference value for the prevention and control of PCV4.
