UPLC-UV-ELSD测定不同产地炒酸枣仁饮片中酸枣仁皂苷A、酸枣仁皂苷B、斯皮诺素和6′′′-阿魏酰斯皮诺素的含量
Content determination of jujuboside A, jujuboside B, spinosin and 6′′′-ferulic acid spinosin in fried Ziziphi Spinosae Semen from different habitats by UPLC-UV-ELSD
【摘要】 目的建立超高效液相色谱(UPLC)-紫外(UV)-蒸发光散射检测法(ELSD)同时测定不同产地炒酸枣仁饮片中酸枣仁皂苷A、酸枣仁皂苷B、斯皮诺素和6′′′-阿魏酰斯皮诺素的含量,对不同产地炒酸枣仁饮片进行质量评价。方法采用Waters Acquity UPLC色谱系统,UV检测器,ELSD检测器,色谱柱为ACQUITY C18柱(150 mm×2.1 mm,1.7μm),流动相为0.1%甲酸水-乙腈(梯度洗脱),流速0.3 mL/min,柱温35℃,UV检测波长335 nm,进样量3μL,喷雾器参数50%,ELSD条件气压172 kPa,漂移管温度60℃。结果在上述色谱条件下测得酸枣仁皂苷A、酸枣仁皂苷B、斯皮诺素和6′′′-阿魏酰斯皮诺素分别在27.20~435.20 mg/mL、12.60~200.80 mg/mL、26.38~422.00 mg/mL、24.38~390.00 mg/mL时与色谱峰面积线性关系良好,平均回收率分别为100.01%、99.99%、100.00%、100.04%,相对标准偏差分别为0.17%、0.32%、0.32%、0.32%。结论 UPLC-UV-ELSD法准确、灵敏,可作为不同产地炒酸枣仁饮片的质量控制方法,测定结果表明,河北产炒酸枣仁饮片中4种成分含量相对较高。
【Abstract】 Objective To establish an ultra-high performance liquid chromatography(UPLC)-ultraviolet(UV)-evaporative light scattering detection(ELSD) method for the determination content of jujuboside A, jujuboside B, spinosin and6′′′-ferulic acid spinosin in fried Ziziphi Spinosae Semen from different habitats,the quality of Fried Ziziphi Spinosae Semen from different habitats was evaluated. Methods The chromatographic separation was achieved on Waters Acquity UPLC system, UV detector, and ELSD detector, performed on ACQUITY C18(150 mm×2.1 mm, 1.7 μm), and the mobile phase was 0.1% formic acid water-acetonitrile(gradient elution), the flow rate was 0.3 m L/min, the column temperature was 35℃, UV the detection wavelength was 335 nm and the injection volume was 3 μL, sprayer parameter was50%, ELSD condition pressure was 172 kPa and drift tube temperature was 60℃. Results Under the above chromatographic conditions, jujuboside A, jujuboside B, spinosin and 6′′′-ferulic acid spinosin had a good linear relationship respectively with the chromatographic peak area at 27.20-435.20 mg/mL、12.60-200.8 mg/mL、26.38-422.00 mg/mL、24.38-390.00 mg/mL,the average recovery were 100.01%、 99.99%、100.00%、100.04% respectively, relative standard deviation were 0.17%,0.32%, 0.32%, 0.32%, respectively. Conclusion UPLC-UV-ELSD method is accurate and sensitive, can be used as a quality control method of fried Ziziphi Spinosae Semen from different habitats. The results of determination show that the contents of the three components in fried Ziziphi Spinosae Semen from Hebei is relatively higher.
