基于TMT技术的柚皮素磷脂复合物抑制大鼠脉络膜新生血管形成的蛋白质组学研究
Proteomics study on inhibition of choroidal neovascularization in experimental animals using naringin phospholipid complex based on TMT technique
【摘要】 目的应用TMT标记结合生物信息学分析技术,研究柚皮素磷脂复合物(NPC对大鼠实验性脉络膜新生血管形成的蛋白质组学的影响。方法建立大鼠脉络膜新生血管模型,随机分为模型组、NPC组和正常对照组,应用TMT标记技术进行蛋白质组学研究,通过检索数据库Gene Ontology Database,对筛查到的差异蛋白进行生物信息学分析。结果获得特有肽段序列10819个,蛋白质2717个。模型组和NPC组、模型组和正常对照组、NPC组和正常对照组,差异表达蛋白分别为113个(上调57个,下调56个)、314个(上调202个,下调112个)、90个(上调22个,下调68个)。通过分析得到这些差异蛋白具有结合作用、抗氧化活性、催化活性、转运活性、转录调节等重要的分子生物学功能。结论基于TMT技术的蛋白质组学方法有助于鉴定出脉络膜新生血管相关的差异蛋白质,为进一步研究柚皮素磷脂复合物抑制实验性脉络膜新生血管的机制提供依据。
【Abstract】 OBJECTIVE To investigate the proteomics of naringin phospholipid complex on choroid neovascularization in experimental animals by using TMT labeling method together with bioinformatics technology. METHODS Choroidal neovascularization model of rats was established and randomly divided into model group, NPC treatment group and normal control group. Pro-teomics was studied through TMT labeling technique. Gene Ontology Database was searched to perform bioinformatics analysis of the screened differential proteins. RESULTS Specific peptide sequences of ten thousand eight hundred and nineteen, proteins of two thousand seven hundred and seventeen were obtained. There were 113(57 up-regulated, 56 down-regulated), 314(202 up-regulated,112 down-regulated) and 90(22 up-regulated, 68 down-regulated) differentially expressed proteins in model group and NPC group, model group and normal control group, NPC group and normal control group, respectively. Through analysis, these differential proteins have important molecular biological functions such as binding, antioxidant activity, catalytic activity,transporter activity and transcription regulator activity. CONCLUSIONS Proteomic approaches based on TMT techniques can help to identify differential proteins associated with choroidal neo-vascularization. It provides evidence for further study on the mechanism of inhibition of experimental choroidal neovascularization by naringin phospholipid complex.
