诊断超声脉冲重复频率对肿瘤化疗局部释药的影响

【摘要】 目的观察相同占空比时,不同脉冲重复频率对阿霉素在肿瘤组织内药物浓度的影响,从而探讨其与超声空化靶向释药的相关性,以期优化超声空化靶向释药的调节参数。方法健康雌性裸鼠40只,建模成功后随机分为4组。阿霉素推注完成后匀速推注稀释的超声造影剂,同时按预定参数进行超声治疗,对照组予以假照。治疗后肿瘤组织取材以高效液相色谱法测定阿霉素药物浓度。结果多组间及两两组间比较阿霉素药物质量浓度差异无统计学意义。结论 PRF为10~50 Hz时肿瘤组织局部药物浓度差异较小,此区间PRF的调整对超声空化靶向释药影响不显著。

【Abstract】 Objective To observe the effect of different pulse repetition frequency on doxorubicin concentration in tumor tissue at the same duty cycle, so as to explore the correlation between doxorubicin and ultrasonic cavitation targeted drug release, in order to optimize the regulation parameters of ultrasonic cavitation targeted drug delivery. Methods 40 healthy female nude mice were randomly divided into four groups after successful modeling. After the completion of doxorubicin injection, the diluted ultrasound contrast agent was injected uniformly, and the ultrasound treatment was carried out according to the predetermined parameters, while the control group was given sham light. After treatment, tumor tissue samples were collected and the concentration of doxorubicin was detected by high performance liquid chromatography. Results There is no significant difference in doxorubicin concentration between multiple groups and between the two groups. Conclusions There was no significant difference in drug concentration in tumor microcirculation when PRF range from 10 Hz to 50 Hz, and the adjustment of PRF in this interval has no significant effect on ultrasound-induced microbubble targeted drug release.
