进展性脑梗死患者外周血中自噬改变的研究

【摘要】 目的研究进展性脑梗死(progressive cerebral infarction,PCI)患者外周血中自噬的改变。方法前瞻性分析2018年10月-2020年10月在我院住院治疗的急性脑梗死患者123例,根据发病48 h内是否发生神经功能恶化分为进展性脑梗死(PCI)组32例和非进展性脑梗死(NPCI)组91例,并选取年龄、性别相匹配的同期健康体检者37例为对照组。分别采用qRT-PCR和ELISA的方法检测3组研究对象外周血中Beclin1、LC3Ⅱ、P62等自噬因子在mRNA和蛋白水平的表达量。结果 (1) PCI组较NPCI组和对照组Beclin1、LC3Ⅱ的mRNA和蛋白表达量增高、P62的mRNA和蛋白表达量降低(均P <0.05);(2) NPCI组较对照组Beclin1、LC3Ⅱ的mRNA和蛋白表达量增高(P <0.05),而P62 mRNA和蛋白的表达量两组间无差异(P> 0.05)。结论 PCI患者早期外周血中存在自噬的过度激活,调控自噬有可能作为潜在的治疗靶点。

【Abstract】 Objective To study the changes of autophagy in peripheral blood of patients with progressive cerebral infarction. Methods 123 patients with acute cerebral infarction admitted to our hospital from October 2018 to October 2020 were divided into PCI group(n=32) and non-PCI group(n=91),according to the incidence of neurological deterioration within 48 hours. 37 cases of age and gender matched control group(the same period of health examination) were selected.The mRNA and protein expressions of Beclin1,LC3 Ⅱ,P62 in the peripheral blood of the three groups were detected by qRT-PCR and ELISA respectively. Results(1) Compared with non-PCI group and control group,the mRNA and protein expression levels of Beclin1 and LC3Ⅱ were higher and the expression level of P62 was lower in PCI group(all P<0.05);(2) The mRNA and protein expression levels of Beclin1 and LC3Ⅱ in non-PCI group were higher than those in control group(P<0.05),but the expression level of P62 had no difference between the two groups(P>0.05). Conclusion Autophagy in peripheral blood of PCI patients is over-activated in the early stage,and regulating autophagy may be a potential therapeutic target.
