植物乳杆菌联合大豆多肽对小鼠肠道菌群的影响

【摘要】 目的为探究植物乳杆菌HZLP-005(Lactobacillus plantarumHZLP-005)联合碱性蛋白酶制备的大豆多肽(soybean peptide prepared by alkaline protease,SPAP)对小鼠肠道菌群的调节作用。方法随机将60只6 w雄性BALB/C小鼠分为4组:分别灌胃生理盐水(K1)、HZLP-005(J2)、生理盐水+2%SPAP(J3)、HZLP-005+2%SPAP(JJ4),菌体浓度109 cfu/ml,每只灌胃体积均为0.1 ml/10 g,连续灌胃30 d后无菌收集粪便,采用高通量测序技术分析各组小鼠肠道菌群多样性与结构组成。结果 JJ4组小鼠肠道菌群多样性及丰富度最好(P<0.01)。在菌群结构组成上,四组样本主要由厚壁菌门和拟杆菌门组成;进一步分析表明,HZLP-005与SPAP联合后对增加乳杆菌属、毛螺旋菌属、瘤胃球菌属、普雷沃氏菌属、Muribaculaceae、等有益菌属的相对丰度,抑制并降低链球菌属、葡萄球菌属、不动细菌属、螺杆菌属、norank_f_Desulfovibrionaceae及肠杆菌属等有害菌属的繁殖,且作用效果优于二者单独作用。结论 HZLP-005联合SPAP能够更有效地提高小鼠肠道菌群的多样性,增加有益菌群丰度,抑制并降低条件致病菌。[营养学报,2021,43(2):191-194,199]

【Abstract】 Objective To explore the effects of HZLP-005 combined with SPAP on the intestinal flora in mice. Method Sixty six-week-old healthy male BALB/C mice were randomly divided into four groups. Mice in the K1 group were gavaged with saline. Mice in the J2 group were gavaged with 109 cfu/ml of HZLP-005. Mice in the J3 group were gavaged with saline supplemented with 2.0% SPAP. Mice in the JJ4 group were gavaged with 109 cfu/ml of HZLP-005 supplemented with 2.0% SPAP. All treatments were lasted for 30 days. The dosage of each gavage was 0.1 ml/10 g. The diversity and structural composition of the intestinal flora were analyzed by high-throughput sequencing technology on the day 30. Results The intestinal flora in the JJ4 group were the most diverse and most abundant(P<0.01=). In terms of bacterial composition, the four groups are mainly composed of Firmicutes and Bacteroides. Further analysis showed that the combination of HZLP-005 and SPAP increased the relative abundance of some beneficial bacteria such as Lactobacillus, Lachnospiraceae, Rumenococcus, Prevotella, Muribculaceae, etc, and inhibited the growth of some harmful bacteria such as Streptococcus, Staphylococcus, Acinetobacter, Helicobacter, norank_f_Desulfovibrionaceae and Enterobacter, etc. The combined effect of HZLP-005 and SPAP was better than those of either alone. Conclusion The HZLP-005 combined with SPAP improved the structure of the intestinal flora through increasing the diversity and the abundance of the dominant flora and inhibited the growth of conditional pathogenic bacteria.
