芦丁和曲克芦丁对吡柔比星诱导大鼠心肌细胞氧化应激损伤的保护作用
PROTECTION OF RUTIN AND TROXERUTIN AGAINST PIRARUBICIN-INDUCED OXIDATIV STRESS INJURY IN RAT CARDIOMYOCYTES
【摘要】 目的探究芦丁(RUT)和曲克芦丁(TRX)对吡柔比星(THP)诱导大鼠心肌细胞(H9C2)氧化应激损伤的保护作用。方法以5μmol/L THP诱导H9C2细胞发生氧化应激反应,不同剂量的RUT和TRX预处理后,MTT法检测细胞存活率,确定最佳保护浓度;流式细胞术检测凋亡细胞数;DCFH-DA荧光探针检测细胞内活性氧(reactive oxygen species,ROS)含量;ELISA试剂盒检测丙二醛(malondialchehyche,MDA)和超氧化物歧化酶(superoxide dismutase,SOD)含量。结果与THP组比较,加入不同浓度的RUT和TRX后,细胞存活率显著提高(P<0.05),并确定40μmol/L RUT和TRX作为最佳保护浓度;流式细胞术结果提示,与THP组相比,RUT和TRX可显著减少凋亡细胞数;与CON组相比,THP组细胞内ROS含量显著增强(P<0.05),与THP组相比,RUT和TRX预处理后细胞内ROS含量均降低(P<0.05);RUT和TRX预处理后细胞内MDA含量显著降低,SOD活性显著升高(P<0.05)。结论 RUT和TRX对THP诱导的氧化应激损伤细胞具有一定的保护作用,其作用机制可能与降低细胞内ROS和MDA含量,提高SOD活性有关。[营养学报,2021,43(1):92-96]
【Abstract】 Objective To investigate the protective effects of rutin (RUT) and troxerutin (TRX) on pirarubicin (THP)-induced oxidative stress injury in rat cardiomyocytes (H9C2).Methods Oxidative stress was induced with 5μmol/L THP in H9C2 cells.After pretreated with different doses of RUT and TRX,the cell survival rate was measured by MTT test to determine the optimal protective concentration.The number of apoptotic cells was measured by flow cytometry.DCFH-DA probe was used to detect reactive oxygen species (ROS) in cells.ELISA kits were used to detect malondialchehyche (MDA)and superoxide dismutase (SOD).Results Compared with the THP group,after RUT and TRX pretreatments,the cell survival rate was significantly improved (P<0.05),and 40μmol/L RUT and TRX were determined as the optimal protective concentration.The results of flow cytometry suggested that compared with the THP group,RUT and TRX could significantly reduce the number of apoptotic cells.Compared with the CON group,the intracellular ROS of the THP group was significantly increased (P<0.05);compared with the THP group,the RUT and TRX pretreatment decreased intracellular ROS(P<0.05).After RUT and TRX pretreatment,intracellular MDA content decreased significantly,and SOD activity increased significantly (P<0.05).Conclusion RUT and TRX have a protective effect on THP-induced oxidative stress in rat cardiomyocytes,and the mechanism is related to reduced content of ROS and MDA in the cell and increased activity of SOD.[ACTA NUTRIMENTA SINICA,2021,43(1):92-96]
