灵芝超微粉对大鼠的长期毒性研究

【摘要】 目的评价灵芝超微粉的临床应用安全性。方法将120只SD大鼠随机分为灵芝超微粉高、中、低剂量组[B组、C组、D组,8.4,4.2,2.1g生药/(kg·24 h)]和溶剂对照组(A组,等体积纯化水),各30只,雌雄各半。灌胃给予相应溶液,每日2次,连续91 d。给药结束后及给药结束30 d后检测大鼠免疫学、尿液、眼科相关指标,以及脏器质量,并计算脏器系数,且进行肉眼观察及组织病理学检查。结果与A组比较,C组雌性大鼠给药结束后的CD4水平显著升高,B组雄性大鼠给药结束后的胸腺质量及系数均显著减少/降低,肾上腺系数显著升高,D组雄性大鼠给药结束后的肝脏系数显著降低(P <0.05)。肉眼观察及组织病理学检查各组大鼠均无明显异常。结论灵芝超微粉长期喂饲大鼠未见明显毒性反应,该研究结果为灵芝超微粉的临床安全应用提供了参考。

【Abstract】 Objective To evaluate the clinical safety of Ganoderma ultra-fine powder. Methods A total of 120 SD rats were randomly divided into Ganoderma ultra-fine powder high-dose,medium-dose and low-dose groups [ group B,group C,group D,8. 4,4. 2,2. 1 g crude drug/( kg·24 h) ] and solvent control group( group A,equal volume of purified water),30 rats in each group,with half male and half female. Rats in each group were given corresponding drugs by gavage twice a day for 91 d. At the end of the administration and30 d after the administration,the relevant indexes of immunology,urine,ophthalmology and organ weight of rats were detected,the organ coefficient was calculated,and the visual observation and histopathological examination were carried out. Results Compared with those in group A,the CD4 level of female rats in group C increased significantly at the end of administration,the thymus weight and coefficient of male rats in group B decreased significantly at the end of administration,the adrenal coefficient of male rats in group B increased significantly at the end of administration,and the liver coefficient of male rats in group D decreased significantly at the end of administration( P < 0. 05). No obvious abnormalities were found in each group by visual observation and histopathological examination.Conclusion Long-term feeding of Ganoderma ultra-fine powder has no obvious toxic reaction in rats,which provides a reference for the safe clinical application of Ganoderma ultra-fine powder.
