非小细胞肺癌组织中PDHA1、GBP1的表达及临床意义
Clinical significance and expression of PDHA1 and GBP1 in non-small cell lung cancer
【摘要】 目的探讨非小细胞肺癌(NSCLC)组织中丙酮酸脱氢酶复合物E1α亚单位(PDHA1)和鸟苷酸结合蛋白1(GBP1)的表达及临床意义。方法选取山东省烟台市烟台山医院2016年1月至2017年10月84例NSCLC患者为研究对象,比较其癌组织及癌旁组织中PDHA1与GBP1的表达情况。根据患者癌组织中PDHA1的表达情况将其分为PDHA1阳性组(30例)和PDHA1阴性组(54例),再根据GBP1的表达情况将其分为GBP1阳性组(58例)和GBP1阴性组(26例)。随访3年,采用生存曲线分析PDHA1与GBP1表达与患者预后的关系。结果 NSCLC患者癌组织中PDHA1表达低于癌旁组织,GBP1表达高于癌旁组织,差异有统计学意义(P <0.05)。84例NSCLC患者均随访成功,无失访病例。随访3年内,共有31例死亡。PDHA1阳性组3年生存率高于PDHA1阴性组,GBP1阳性组3年生存率低于GBP1阴性组,差异有统计学意义(P <0.05)。结论 PDHA1在NSCLC组织中低表达,GBP1在其中高表达,二者有望成为新的NSCLC预后评估的分子标志物。
【Abstract】 Objective To investigate the expression and clinical significance of pyruvate dehydrogenase complex E1αsubunit(PDHA1) and guanylate binding protein 1(GBP1) in non-small cell lung cancer(NSCLC). Methods A total of84 patients with NSCLC were selected from Yantaishan Hospital of Yantai City, Shandong Province from January 2016 to October 2017. The expressions of PDHA1 and GBP1 in the cancerous and adjacent tissues were compared. The patients were divided into PDHA1 positive group(30 cases) and PDHA1 negative group(54 cases) according to the expression of PDHA1 in cancer tissues, and then divided into GBP1 positive group(58 cases) and GBP1 negative group(26 cases)according to the expression of GBP1. The patients were followed up for three years, and the relationship between PDHA1 and GBP1 expression and prognosis was analyzed by survival curve. Results The expression of PDHA1 in the cancerous tissues of NSCLC patients was lower than that in the adjacent tissues, and the expression of GBP1 was higher than that in the adjacent tissues, with statistical significance(P < 0.05). All 84 patients with NSCLC were followed up successfully, and no cases were lost to follow-up. During three years of follow-up, 31 cases died. The 3-year survival rate of PDHA1 positive group was higher than that of PDHA1 negative group, and the 3-year survival rate of GBP1 positive group was lower than that of GBP1 negative group, with statistical significance(P < 0.05). Conclusion Low expression of PDHA1 and high expression of GBP1 in NSCLC tissues are expected to become new molecular markers for prognosis assessment of NSCLC.
