【摘要】 目的探究血管紧张素转换酶2(angiotensin-converting enzyme 2,ACE2)在卵巢癌的表达情况及其与预后的相关性。方法通过HPA、cBioPortal数据库分析ACE2在恶性肿瘤的表达谱,结合GEPIA、Oncomine数据库进一步探究ACE2在卵巢癌的表达情况,通过生存分析和药物敏感性分析评估ACE2表达与卵巢癌预后的相关性。结果卵巢癌是常见的表达ACE2的恶性肿瘤,约20%的卵巢癌患者表达ACE2。4%的卵巢癌患者有ACE2突变,以扩增突变最为常见。在浆液性卵巢癌中,ACE2突变以扩增突变为主且有拷贝数增加,但其表达水平低于其他卵巢癌亚型。对比分析发现,卵巢癌组织ACE2表达水平高于正常卵巢组织。ACE2高表达卵巢癌患者无进展生存率和总生存率均高于ACE2低表达者(P<0.01)。紫杉醇敏感的卵巢癌细胞系ACE2表达水平最高,紫杉醇耐受的卵巢癌细胞系ACE2表达水平最低。结论 ACE2在卵巢癌中较为常见,常有扩增突变。ACE2高表达有利于卵巢癌患者的预后。更多还原

【Abstract】 Objective　To study the expression of angiotensin-converting enzyme 2 (ACE2) in ovarian cancer and its relationship with prognosis.Method The expression profile of ACE2 in malignant tumors was analyzed by HPA and cBioPortal database,and the expression of ACE2 in ovarian cancer was further explored in combination with GEPIA and Oncomine database.The correlation between ACE2 expression and prognosis of ovarian cancer was evaluated by survival analysis and drug sensitivity analysis.Result Ovariancancer was one of the most common malignant tumor that expresses ACE2.About 20% of ovarian cancer patients express ACE2.4% of ovarian cancer patients had ACE2 mutations,and amplification was the most common.In serous ovarian cancer,ACE2 mutations were mainly amplification with increased copy number,but its expression level was lower than other ovarian cancer subtypes.Comparative analysis found that the expression level of ACE2 in ovarian cancer tissue was higher than that in normal ovarian tissue.The progression-free survival rate and overall survival rate of ovarian cancer patients with high ACE2 expression were higher than those with low ACE2 expression (all P<0.01).Paclitaxel-sensitive ovarian cancer cell lines had the highest expression levels of ACE2,while paclitaxel-resistant ovarian cancer cell lines had the lowest expression levels of ACE2.Conclusion ACE2 is common in ovarian cancer with amplification mutations.High expression of ACE2 promotes the prognosis of patients with ovarian cancer.更多还原
