高效液相色谱一测多评法测定酸枣仁中的2个黄酮活性成分含量

【摘要】 目的:建立酸枣仁中2个黄酮活性成分的高效液相色谱一测多评法。方法:以斯皮诺素为内参物,通过斜率法确定6′′′-阿魏酰斯皮诺素的相对校正因子,再用相对保留值法对6′′′-阿魏酰斯皮诺素的色谱峰进行定位。最后,采用所建立的一测多评法对29批酸枣仁药材中2个黄酮活性成分进行含量测定,同时与外标法测得的含量结果进行比较。结果:6′′′-阿魏酰斯皮诺素的相对校正因子为0.73,相对保留时间值为1.84。一测多评法和外标法所得含量测定结果无显著性差异。结论:本法能准确有效地控制酸枣仁中黄酮类成分的内在质量,为其药典标准的完善提供技术依据。

【Abstract】 Objective: To establish a method of "Quantitative analysis of multi-components by singlemarker(QAMS)" with high performance liquid chromatography to control the quality of Ziziphi Spinosae Semen by determining its two active components of flavonoids. Methods: Using spinosin as the internal reference substance, the relative correction factor of 6′′′-feruloylspinosin was determined by the ratio of slopes. Then the relative retention time was used to locate the peak of 6′′′-feruloylspinosin. Finally, the contents of two flavonoids in 29 batches of Ziziphi Spinosae Semen were determined by the established QAMS method comparing with those determined by the external standard method. Results: The relative correction factor of 6′′′-feruloylspinosin was determined as 0.73, and its relative retention time was 1.84.There was no significant difference in content results between these two methods. Conclusions: The established QAMS method can be used in the quality control of Ziziphi Spinosae Semen, which provides technical basis for the improvement of the pharmacopoeia standard of Ziziphi Spinosae Semen.
