NF-κB和iNOS在ALT≥400U/L慢性乙型肝炎患者的表达及临床意义
The Expression and Clinical Significance of NF-κB and iNOS in Patients with Chronic Hepatitis B(ALT≥400U/L)
【摘要】 目的:研究慢性乙型肝炎患者外周血中核因子-κB(Nuclear factor-κB,NF-κB)、诱导型一氧化氮合成酶(inducible nitric oxide synthase, iNOS)的表达情况,分析二者的临床价值。方法:纳入2017年4月10日—2020年4月10日于我院住院的慢性乙型肝炎患者80例作为肝炎组,60例健康者作为对照组。采用RT-PCR法检测NF-κB mRNA和iNOS mRNA的表达;Western Blot法检测外周血中NF-κB、iNOS蛋白的表达;ELISA法检测血清中NO水平。结果:(1) RT-PCR法检测结果发现:肝炎组NF-κB mRNA和iNOS mRNA的表达水平均高于对照组(均P<0.05);(2) Western Blot法检测结果发现:肝炎组NF-κB和iNOS蛋白表达水平均高于对照组(均P<0.05);(3)肝炎组NO水平高于对照组(P<0.05)。结论:本研究发现CHB患者NF-κB、iNOS表达水平较健康组明显升高,且分泌大量炎症因子NO,在HBV感染的致炎过程中发挥重要作用。
【Abstract】 Objective:To study the expression of nuclear factor-κB(NF-κB) and inducible nitric oxide synthase(iNOS) in peripheral blood of patients with chronic hepatitis B,and analyze their clinical value.Methods:A total of 80 patients with chronic hepatitis B who were hospitalized in our hospital from April 10 th 2017 to April 10 th 2020 were included as the hepatitis group, and 60 healthy peoples were included as the control group.RT-PCR method was used to detect the expression of NF-κB mRNA and iNOS mRNA;Western Blot method was used to detect the expression of NF-κB and iNOS protein in peripheral blood; ELISA method was used to detect the level of NO in serum.Results:(1)The results of RT-PCR method showed that: the expression levels of NF-κB mRNA and iNOS mRNA in the hepatitis group were significantly higher than the control group(both P<0.05);(2)The Western Blot method showed that: the expression levels of NF-κB and iNOS protein in hepatitis group were significantly higher than control group(all P<0.05);(3)The NO level of hepatitis group was significantly higher than that of the control group(P<0.05).Conclusion:This study found that the expression levels of NF-κB and iNOS in CHB patients were significantly higher than those in the healthy group, and they secreted a large amount of inflammatory factor NO,which played an important role in the inflammation process of HBV infection.
