七种独活、当归药材中蛇床子素及总香豆素含量比较
Comparison of Osthole and Total Coumarin Contents in Seven Different Kinds of Radix Angelicae and Angelica Pubescens
【摘要】 目的:测定川独活、贵州野生独活、湖北肉独活、四川牛尾独活、四川九眼独活以及亲缘关系相近的当归、毛当归中的蛇床子素、总香豆素含量。方法:采用高效液相色谱法测定药材中蛇床子素含量,色谱柱选择Hypersil BDS C18柱(4.6mm×250mm),流动相为水(A)-甲醇(B),二元梯度洗脱(0~6min时60%B,6~12min时60%B→50%B,12~20min 50%B→45%B,20~35min 45%B→60%B),流速1.0mL/min,柱温30℃,进样量20μL,检测波长322nm。采用紫外分光光度法测定药材中总香豆素含量,检测波长322nm。结果:蛇床子素的线性关系方程为:Y=6.651 2×105 X-17.11(R2=0.999 9),线性范围1.375~22μg/mL。总香豆素的线性关系方程为:Y=0.050 5 X+0.064 6(R2=0.997),线性范围1.6~16μg/mL。其中,以贵州野生独活、湖北肉独活中蛇床子素的含量最高,分别为1.02mg/g、0.65mg/g;当归、毛当归中蛇床子素含量为零。以贵州野生独活、毛当归中总香豆素的含量最高,分别为54.85mg/g、48.52mg/g;当归中含量最低为16.73mg/g。结论:不同品种独活中蛇床子素及总香豆素含量存在一定差异,其中贵州野生独活质量明显优于四川、湖北所产品种。毛当归药材中不含蛇床子素,但总香豆素含量很高。采用高效液相色谱法、紫外分光光度法测定独活、毛当归药材中有效成分的含量,方法操作简单,重复性好,精密度高,是不同品种独活中药材质量控制的科学检测方法。
【Abstract】 Objective:To determine the content of osthole and total coumarin in different Radix Angelicae and Angelica Pubescens.Methods:The content of osthole in medicinal materials was determined by HPLC.The chromatographic column was Hypersil BDS C18 column(4.6 mm×250 mm),the mobile phase was water(A)-methanol(B),binary gradient elution(60%B in 0~6 min,60%B→50%B in 6~12 min,50%B→45%B in 12~20 min,45%B→60%B in 20~35 min).The flow rate was 1.0 mL/min,the column temperature was 30℃,and the injection volume was 20μL.The content of total coumarin in medicinal materials was determined by ultraviolet spectrophotometry with detection wavelength of 322 nm.Results:The linear equation of osthole was Y=6.651 2×105 X-17.11(R2=0.999 9),and the linear range was 1.375~22μg/mL.The linear equation of total coumarin was Y=0.050 5 X+0.064 6(R2=0.997),and the linear range was 1.6~16μg/mL.Among them,the content of osthole in Guizhou and Hubei is the highest,which is 1.02 mg/g and 0.65 mg/g respectively;the content of Angelica sinensis and Angelica pubescens is zero.The content of total coumarin in Guizhou and Angelicae Pubescens is the highest,which can reach 54.85 mg/g and 48.52 mg/g respectively.The lowest content in Angelica sinensis is 16.73 mg/g.Conclusion:There are some differences in the content of osthole and total coumarin in different varieties of Radix Angelicae Sinensis Genus Plants,among which the quality of wild Radix Angelicae Pubescentis in Guizhou is obviously better than that in Sichuan and Hubei.There is no osthole in Angelicae Pubescens,but the content of total coumarin is very high.The content of effective components in different Radix Angelicae and Angelica Pubescens was determined by HPLC and UV spectrophotometry.The method is simple in operation,good in repeatability and high in precision.At the same time,it provides a scientific detection method for the quality control.
