貉源大肠杆菌毒力基因的检测与分析
Detection of Virulence Genes in Escherichia coli Isolated from Racoon Dog
【摘要】 为了研究中国东北地区貉(Nyctereutes procyonoides)大肠杆菌的流行情况,对56株源自东北地区不同貉养殖场的貉源大肠杆菌进行主要毒力基因的检测与分析,测定其阳性率与分布情况。利用PCR技术法检测貉源大肠杆菌的iucD,irp2,fyuA,fimH,iss,stx1,ybtA,irp1,stx2,eae 10个毒力基因。结果显示iucD,irp2,fyuA,fimH,ybtA的阳性率分别为39.29%(22/56),37.50%(21/56),53.57%(30/56),98.21%(55/56),5.36%(3/56),而iss,stx1,irp1,stx2,eae均未检出。黏附相关基因fimH及HPI毒力岛核心基因fyuA检出率最高,说明此次试验中fimH及fyuA为貉源大肠杆菌的主要毒力基因。
【Abstract】 To study the prevalence of Escherichia coli in raccoon dog( Nyctereutes procyonoides)from northeastern China we measures the positive rates and distribution,and tested 56 strains of E. coli in raccoon dog from farms in northeastern China. We tested ten virulence genes of E. coli,viz. iucD,irp2,fyuA,fimH,iss,stx1,ybtA,irp1,stx2 and eae by polymerase chain reaction( PCR). The positive rates of iuc D,irp2,fyuA,fimH and ybtA were 39%( 22/56),38%( 21/56),54%( 30/56),98%( 55/56),5%( 3/56),respectively,while iss,stx1,irp1,stx2 and eae were negative. We found that the adhesion-associated gene fimH and the high pathogenicity island-core gene fyuA had high detection rates. For this experiment,fimH and fyuA were the main virulence genes of raccoon dog-origin E. coli.
