思茅藤不同极性部分化学成分分析及其免疫抑制活性
Chemical composition and immunosuppressive activity of different fractions from Epigynum auritum
【摘要】 探究了思茅藤提取物不同极性部分的主要化学成分及其免疫抑制活性情况,并对其可能的活性机制进行初步探讨.利用D101大孔吸附树脂对思茅藤提取物不同极性部分进行划分,采用超高效液相色谱-高分辨质谱联用技术(UHPLC-MS/MS)分析各提取物的主要化学成分,并建立刀豆蛋白A(Con A)和脂多糖(LPS)刺激的小鼠脾淋巴细胞模型,通过测定不同极性部分的思茅藤提取物对小鼠脾淋巴细胞增殖及相关细胞因子分泌的影响评价其免疫抑制活性.结果显示,思茅藤提取物Fr.20%~40%段的化学成分主要为酚类物质,而Fr.80%~100%段富含以甾体类化合物为主的成分.Fr.100%部分能有效的减少Con A刺激的过量炎症因子的产生,显示出最强的免疫抑制活性,且该抑制效果主要发生在G0/G1期.
【Abstract】 This study aimed to comparatively investigate the Chemical composition and immunosuppressive activity of different fractions from Epigynum auritum and its mechanism. The E. auritum extract(EAE) was fractionated by column chromatography with macroporous resin D101. UHPLC-MS/MS was used to analyze the chemical composition of different fractions of EAE. The immunosuppressive effect of different fractions of EAE were evaluated on Con A and LPS-induced mice spleen cells, and the cell proliferation and contents TNF-α and IL-6 were recorded. The major chemical composition of Fr.20%~40% of EAE are polyphenols, while the Fr.80%~100% fractions are rich in steroidal compounds. Fr.100% showed the strongest immunosuppressive effect, inhibited the productions of TNF-a and IL-6 in Con A induced T cells and the cell cycle arrested in in the G0/G1 phase.
