2,4-二硝基苯肼衍生化共振散射光谱法测定丙二醛
2,4-Dinitrophenylhydrazine Derivatization Resonance Scattering Spectrometry for Detection of Malonaldehyde
【摘要】 在pH3.8醋酸-醋酸钠缓冲溶液中,丙二醛(MDA)与2,4-二硝基苯肼(DNPH)发生反应生成疏水性的苯腙,导致体系产生共振散射效应,体系在451 nm、493 nm处产生共振散射峰,据此建立一种用共振散射光谱测定MDA含量的方法。探讨缓冲溶液pH值及其用量、2,4-二硝基苯肼稀乙醇溶液用量和反应时间、反应温度对体系的影响,优化实验条件。在最佳实验条件下,方法的线性范围为0.075~2.5μg/m L,相关系数(R2)为0.9929,检出限(3S/K)为0.025μg/m L。
【Abstract】 In acetic acid sodium acetate buffer solution,pH 3. 8,malonaldehyde( MDA) react with 2,4-dinitrophenylhydrazine to form hydrophobic phenylhydrazone. The system has strong resonance scattering( RS) effect at 451 nm、493 nm. Accordingly,a resonance scattering spectrometry for the determination of MDA is established. Conditions that would affect the systems RS value are studied in detail,including p H,dosages of buffer solution,concentration of 2,4-dinitrophenylhydrazine and reaction time and temperature. Under the optimal condition,the calibration curve is linear within the range of 0. 075 μg/m L-2. 5 μg/m L with the correlation coefficient of 0. 9929,the detection( 3 S/K) limit of0. 025 μg/m L.
