直肠癌组织中microRNA-29a的表达水平及临床意义
Expression level and clinical significance of microRNA-29a in rectal cancer tissues
【摘要】 目的研究直肠癌组织中microRNA-29a的表达水平及临床意义。方法选取2019年3月至2020年10月本院收治的60例直肠癌患者作为研究对象。通过qPCR试剂盒检测其直肠癌组织中microRNA-29a的相对表达量,分析其与临床病理特征的关系,再采用多因素非条件Logistic回归分析探讨影响microRNA-29a相对表达量的相关因素。取直肠癌细胞进行细胞增殖、迁移、侵袭实验,根据反义寡核苷酸(ASO)序列不同将其分为ASO-microRNA-29a组、ASO-对照序列组和空白对照组,比较各组细胞在不同时间点的增殖、迁移、侵袭能力。结果 TNM分期为Ⅲ、Ⅳ期及淋巴结转移患者的microRNA-29a相对表达量均明显高于TNM分期为Ⅰ、Ⅱ期及无淋巴结转移的患者(P<0.05)。多因素非条件Logistic回归分析结果显示,TNM分期为Ⅲ、Ⅳ期及淋巴结转移均为影响直肠癌组织中microRNA-29a相对表达量的相关因素(OR=1.747、2.002,P<0.05)。与ASO-对照序列组、空白对照组比较,ASO-microRNA-29a组转染12、24、48、72、96 h的microRNA-29a增殖能力均降低,迁移及侵袭细胞数均减少(P<0.05)。结论 MicroRNA-29a在直肠癌组织中呈现出高表达,通过阻碍其表达或可抑制肿瘤细胞的增殖、迁移与侵袭能力。
【Abstract】 Objective To investigate the expression level and clinical significance of microRNA-29 a in rectal cancer tissues. Methods Sixty patients with rectal cancer treated in our hospital from March 2019 to October 2020 were selected as the research objects. The relative expression of microRNA-29 a in rectal cancer tissues was detected by qPCR kit, its relationship with clinicopathological features was analyzed, and then the related factors affecting the relative expression of microRNA-29 a were discussed by multivariate unconditional Logistic regression analysis. Rectal cancer cells were taken for cell proliferation, migration and invasion experiments. According to different antisense oligonucleotide(ASO)sequences, they were divided into ASO-microRNA-29 a group, ASO control sequence group and blank control group. The proliferation, migration and invasion abilities of cells in each group were compared at different time points. Results The relative expression of microRNA-29 a in patients with TNM stage Ⅲ and Ⅳ and lymph node metastasis were significantly higher than those in patients with TNM stage Ⅰ and Ⅱ and no lymph node metastasis(P <0.05). Multivariate unconditional Logistic regression analysis result showed that TNM stage Ⅲ and Ⅳ and lymph node metastasis were related factors affecting the relative expression of microRNA-29 a in rectal cancer(OR=1.747, 2.002, P<0.05). Compared with the ASO control sequence group and blank control group, the proliferation ability of microRNA-29 a transfected for12, 24, 48, 72 and 96 h in the ASO-microRNA-29 a group decreased, and the number of migrating and invasive cells decreased(P <0.05). Conclusion MicroRNA-29 a is highly expressed in rectal cancer tissue, which may inhibit the proliferation, migration and invasion abilities of tumor cells by blocking its expression.
