五味子乙素改善酒精中毒大鼠学习记忆能力的作用研究

【摘要】 目的探讨五味子乙素(Schisandrin B,Sch B)对酒精中毒大鼠学习记忆能力的影响及神经细胞保护作用。方法每日用56°北京红星二锅头白酒(8ml/kg)连续灌胃8周制备慢性酒精中毒大鼠模型,造模成功后Sch B低剂量组、Sch B中剂量组、Sch B高剂量组再继续给予不同剂量的Sch B灌胃30d。然后用Morris水迷宫检测大鼠的定位航行实验训练潜伏期和游泳路径轨迹图,透射电镜和光镜观察海马神经细胞形态结构的改变。结果 Morris水迷宫结果显示模型组大鼠潜伏期长(P <0.05),从运动轨迹图显示,Sch B各浓度组运动趋向目标象限明确;光镜和透射电镜观察到Sch B各浓度组大鼠海马CA1区神经细胞形态较为规则,细胞膜较为完整,神经细胞水肿减轻,神经元损伤数量和损伤程度较酒精中毒模型组有明显减少。结论 Sch B能改善慢性酒精中毒大鼠的学习记忆能力,对海马组织神经细胞具有保护作用。

【Abstract】 Objective To investigate the effect of Schisandrin B(Sch B)on the learning and memory ability and its protective effect on nerve cells of rats with chronic alcoholism.Methods The rat models with chronic alcoholism were established by continuous intragastric administration of 56°Beijing Red Star Erguotou Liquor(8 ml/kg)for 8 weeks.After successful modeling,the low-dose Sch B group,the medium-dose Sch B group and the high-dose Sch B group were given different doses of Sch B by intragastric administration for 30 days.Then Morris water maze was used to detect the experiment training latency of the rats’ positioning navigation and their swimming path trajectory.The morphological and structural changes of hippocampal neurons were observed by transmission electron microscope and light microscope.Results The Morris water maze results showed that the latency of rats in the model group was longer(P <0.05).The movement trajectory chart indicated that the movement trend of each Sch B group was clear in the target quadrant.Light microscope and transmission electron microscopy showed that the morphology of neurons in the hippocampal CA1 area of rats in each Sch B group was regular,with complete cell membranes.In comparison with the alcoholism group,the edema of neurons alleviated,the number of damaged neurons and their damaging degree significantly decreased in the Sch B groups.Conclusion Sch B can improve the learning and memory ability of rats with chronic alcoholism and protect the neurons in hippocampus.
