黄腐醇对肺癌细胞株A549体外抑制作用及其可能机制

【摘要】 目的探讨黄腐醇对人肺癌细胞株A549的体外抑制作用及其可能机制。方法体外培养A549细胞,采用MTT法检测黄腐醇10、20μmol/L或联用N-乙酰-L-半胱氨酸(NAC)5 mmol/L对细胞生长的作用,细胞集落实验检测细胞的集落形成能力,流式细胞术检测细胞凋亡情况和活性氧(ROS)含量,Western blot法检测p53、p21、转录激活因子4(ATF4)和CCAAT/增强子结合蛋白同源蛋白(CHOP)的蛋白表达。结果黄腐醇呈浓度和时间依赖性地抑制A549细胞生长(P<0.05)。与对照组相比,黄腐醇10、20μmol/L均可减少A549细胞集落的数量,促进细胞凋亡,增加细胞内ROS含量以及p53、p21、ATF4、CHOP蛋白表达(P<0.05);而NAC预处理后,可降低黄腐醇诱导的ROS含量以及细胞抑制率(P<0.05)。结论黄腐醇能够体外抑制肺癌细胞生长并促进细胞凋亡,可能与黄腐醇升高肿瘤细胞内ROS含量、调节细胞周期和内质网应激相关蛋白的表达有关。

【Abstract】 Objective To investigate the inhibitory effect and underlying mechanism of xanthohumol on lung cancer cell line A549 in vitro.Methods A549 cells were culture in vitro,and the effect of xanthohumol 10,20 μmol/L or combination with N-Acetyl-L-cysteine(NAC) 5 mmol/L on the cell growth was measured by MTT method.The colony forming ability of the cells was determined by cell colony assay.The cell apoptosis and reactive oxygen species(ROS) production were analyzed by flow cytometry.The protein expressions of p53,p21,activating transcription factor 4(ATF4) and CCAAT/enhancer binding protein homologous protein(CHOP) were detected by Western blot.Results Xanthohumol could inhibit the growth of A549 cells in a dose-and time-dependent manner(P<0.05).Compared with control group,xanthohumol 10,20 μmol/L decreased the number of cell colonies,promoted cell apoptosis,and increased ROS production and protein expressions of p53,p21,ATF4 and CHOP(P<0.05).While the pretreatment with NAC could decrease xanthohumol-induced ROS production and cell inhibition rate(P<0.05).Conclusion Xanthohumol can inhibit cell growth and promote cell apoptosis of lung cancer in vitro,which may be related to the increase of ROS production in cancer cells,regulation of cell cycle and endoplasmic reticulum stress-related protein expressions.
