贯叶连翘不定根不同溶剂提取物的抗氧化活性研究
Antioxidant activity of different solvent extracts from adventitious roots of Hypericum perforatum
【摘要】 为探究生物反应器培养的贯叶连翘不定根抗氧化活性,该研究用正丁醇、甲醇、乙醇和水4种溶剂对不定根进行提取,研究了不同溶剂提取物的DPPH自由基清除能力、ABTS自由基清除能力和铁离子螯合能力,并通过层次分析法对抗氧化能力进行综合评价。结果表明:4种溶剂提取物中水提物的抗氧化能力很弱,甲醇、乙醇和正丁醇提取物的抗氧化能力随提取物浓度的升高而增强,其中甲醇提取物对DPPH自由基清除活性最强,乙醇提取物对ABTS自由基清除活性最强,而正丁醇提取物的铁离子螯合能力最强。利用3种抗氧化评价中的半数有效浓度进行了层次分析,得到正丁醇、甲醇、乙醇提取物的综合评分值分别为1.469、1.360和0.975,表明乙醇提取物的抗氧化综合能力最强。因此,贯叶连翘不定根乙醇提取物可用抗氧化相关产品生产中。
【Abstract】 To explore antioxidant activity of bioreactor-cultured adventitious roots(ARs) of Hypericum perforatum, the ARs were extracted using four different solvents(i.e., n-butanol, methanol, ethanol and water) and antioxidant activities of extracts were investigated using three evaluating methods, namely, DPPH free radical scavenging ability, ABTS radical scavenging ability, and the Fe2+ chelating ability. Finally, the half maximal effective concentration of n-butanol, methanolic, and ethanolic extracts from different methods of antioxidant evaluation was as the index to comprehensively evaluate the antioxidant ability of different extracts through an analytic hierarchy process(AHP). The results showed that antioxidant activity of water-extract was low and those of methanolic, ethanolic, and n-butanol extracts increased with extract concentration increased. The highest value of DPPH radical scavenging ability was found on the methanolic extract, and those of ABTS radical scavenging capacity on the ethanolic extract and Fe2+ chelating ability on the n-butanol extract. The result of AHP showed that the comprehensive scores of n-butanol, methanolic, and ethanolic extracts were 1.469, 1.360, and 0.975, respectively, indicating that ethanolic extract had the strongest antioxidant capacity. Therefore, ethanolic extract of H. perforatum ARs could be used in production of antioxidant products.
