食管癌细胞血管生成拟态的形态学及机制研究

【摘要】 目的通过体外培养对食管癌细胞血管生成拟态进行形态学研究,并探讨其可能的分子机制。方法体外培养食管癌细胞Eca-109、Kyse-150、TE-10,观察其是否具有形成血管生成拟态的能力。体外培养Eca-109细胞,分别用0、10、20、30μmol/L的通路抑制剂LY294002处理细胞24h,应用MTT法检测LY294002对细胞增殖及形成血管生成拟态能力的影响。Western blotting法检测LY294002对Eca-109细胞Akt及pAkt蛋白表达的影响。结果食管癌细胞在体外培养下能够形成血管生成拟态,Eca-109细胞血管生成拟态形成数量最多,与Kyse-150细胞相比,差异有统计学意义(P<0.05)。LY294002对食管癌Eca-109细胞增殖有抑制作用,且呈剂量依赖性。Eca-109细胞加入不同浓度的LY294002处理24h后,血管生成拟态形成能力有下降的趋势,浓度为30μmol/L时,抑制作用最为明显(P<0.05)。Western blotting结果显示,不同浓度LY294002对食管癌Eca-109细胞Akt蛋白表达水平的影响无统计学差异(F=3.78,P>0.05),对食管癌Eca-109细胞pAkt蛋白表达水平的影响有统计学差异(F=33.23,P<0.05)。结论食管癌细胞在体外培养条件下均具有形成血管生成拟态的能力,不同来源的食管癌细胞,血管生成拟态形成能力不同。PI3K/Akt通路参与食管癌细胞血管生成拟态的形成。

【Abstract】 Objective To research the morphology of esophageal cancer cells vasculogenic mimicry and investigate its possible molecular mechanism.Methods The ability of vasculogenic mimicry in esophageal cancer cells Eca-109,Kyse-150 and TE-10 of cultured in vitro were observed.Eca-109 cells were cultured in vitro and treated with LY294002(0,10,20,30μmol/L)for 24 h,and the influences of LY294002 on cell proliferation and vasculogenic mimicry ability were detected by MTT assay.The influences of pathway inhibitor LY294002 on expressions of Akt and pAkt protein in Eca-109 cells were detected by Western blotting.Results Esophageal cancer cells could form vasculogenic mimicry in vitro culture,and the number of vasculogenic mimicry in Eca-109 cells was more than Kyse-150 cells,the difference was statistically significant(P<0.05).The LY294002 inhibited the proliferation of Eca-109 cells in a dose-dependent manner.The ability of vasculogenic mimicry was decreased when the Eca-109 cells were treated by different concentrations of LY294002 for 24 h,and the inhibition effect was most obvious when the concentration was 30μmol/L(P<0.05).Western blotting showed there were no significant differences of Akt protein expression among different concentrations of LY294002(F=3.78,P>0.05),but there were significant differences of pAkt protein expression among different concentrations of LY294002(F=33.23,P<0.05).Conclusions Esophageal cancer cells have the ability to form vasculogenic mimicry in vitro culture.Due to the different origin of esophageal cancer cells,the ability of vasculogenic mimicry is different.PI3 K/Akt pathway involves in the formation of vasculogenic mimicry in esophageal cancer cells.
