【摘要】 目的:分析食管癌组织中miR-503和钙黏蛋白（E-cadherin）的表达情况,探讨miR-503、E-cadherin与食管癌患者预后的关系。方法:选取2013-09—2015-03宝鸡市中心医院胸外科接受外科手术治疗的119例食管癌患者,术中收集相应的食管癌组织和癌旁正常组织。采用实时荧光定量PCR法检测组织中miR-503的相对表达量;采用免疫组化法检测食管癌组织中E-cadherin表达水平;采用Pearson法分析食管癌组织中miR-503表达水平与E-cadherin表达积分的相关性;对所有患者术后进行随访,采用Kaplan-Meier法分析食管癌组织中miR-503和E-cadherin表达与患者5年累积生存率的关系。结果:与癌旁正常组织比较,食管癌组织中miR-503相对表达量显著上升,E-cadherin阳性表达率、表达积分显著降低（P<0.05）。与浸润深度T₁+T₂、TNM分期Ⅰ+Ⅱ、高分化、无淋巴结转移患者比较,浸润深度T₃+T₄、TNM分期Ⅲ+Ⅳ、中和低分化、有淋巴结转移的患者miR-503相对表达量显著上升,而E-cadherin表达积分显著降低（P<0.05）。食管癌组织中miR-503表达水平与E-cadherin表达积分呈负相关（r=-0.517,P<0.05）。食管癌组织中miR-503高表达者5年累积生存率低于miR-503低表达者（42.31%vs 65.85%,χ²=4.962,P<0.01）,E-cadherin高表达者5年累积生存率高于E-cadherin低表达者的（79.59%vs 30.00%,χ²=25.803,P<0.01）。结论:miR-503在食管癌组织中呈高表达,E-cadherin呈低表达,二者与食管癌的发生及患者预后密切相关。更多还原

【Abstract】 Objective:To detect the expressions of miR-503 and E-cadherin in esophageal carcinoma,and to explore the correlation between miR-503,E-cadherin and prognosis of esophageal cancer.Method:From September2013 to March 2015,119 patients with esophageal cancer who accepted surgical resection in the department of thoracic Surgery of Baoji Central Hospital were selected,the corresponding esophageal cancer tissues and adjacent normal tissues were collected during the operation.The relative expression of miR-503 in tissues was detected by realtime fluorescent quantitative PCR;the expression level of E-cadherin was detected by immunohistochemistry;Pearson method was used to analyze the correlation between the expressions of miR-503 and E-cadherin in esophageal cancer tissues;in addition,all patients were followed up,and Kaplan-Meier method was used to analyze the relationship between the expressions of miR-503 and E-cadherin and 5-year cumulative survival rate in esophageal cancer.Results:Compared with adjacent normal tissues,the relative expression of miR-503 was increased significantly,and the positive expression rate and expression score of E-cadherin were decreased significantly（P<0.05）.Compared with infiltration depth T₁+ T₂,TNM stageⅠ + Ⅱ,high differentiation and no lymph node metastasis,the relative expression of miR-503 was increased significantly in patients with infiltration depth T₃+ T₄,TNM stageⅢ+ Ⅳ,medium and low differentiation and lymph node metastasis,while the expression of E-cadherin was decreased significantly（P<0.05）.There was a negative correlation between the expression of miR-503 and E-cadherin in esophageal carcinoma（r=-0.517,P<0.05）.The 5-year cumulative survival rate of patients with high expression of miR-503 was lower than patients with low expression of miR-503（42.31% vs 65.85%,χ²=4.962,P<0.01）.The 5-year cumulative survival rate of patients with high expression of E-cadherin was higher than patients with low expression of E-cadherin（79.59% vs 30.00%,χ²=25.803,P<0.01）.Conclusion:The expression of miR-503 is high in esophageal carcinoma and the expression of E-cadherin is low;moreover,they are closely related to the occurrence and prognosis of esophageal cancer.更多还原
