低纤维饮食联合亚甲蓝肛门注射法建立功能性排便障碍动物模型
Low-fiber diet combined with methylene blue anal injection to establish an animal model of functional defecation disorder
【摘要】 目的探讨采用低纤维饮食联合亚甲蓝肛门注射法建立功能性排便障碍(function defecation disorder, FDD)动物模型。方法低纤维饮食联合局部亚甲蓝肛门注射法建立FDD动物模型,通过对大鼠粪便性状、粪便质量、胃肠传输功能、排便功能、大鼠行为学、结肠及直肠组织形态学、血清及直肠组织中脑肠肽含量进行观察,以评估FDD模型构建情况。结果亚甲蓝各剂量组粪便干硬,呈黑褐色,结、直肠固有层可见大量的炎性细胞浸润,伴见血管扩张,球囊排出时间延长,肛管直肠静息压降低,开臂滞留时间百分比(OT%)及开臂进入次数百分比(OE%)降低,血清及直肠组织中脑肠肽含量水平异常,胃肠传输功能无明显改变。结论低纤维饮食联合亚甲蓝肛门注射法建立的大鼠动物模型具有FDD的病理特征,可作为一个较理想的动物模型进一步用于药物治疗学及发病机制的研究。
【Abstract】 Objective To establish the animal model of functional defecation disorder(FDD) by low-fiber diet combined with perianal area injection of methylene blue. Methods A low-fiber diet combined with local perianal injection methylene blue was used to establish an animal model of FDD. To evaluate the construction of FDD animal model, the fecal characteristics, fecal quality, gastrointestinal transit function, defecation function, behavioral experiment, colon and rectum histomorphology, serum and colon tissue brain-gut peptide content of rats were observed. Results The methylene blue injection group had dry and hard stools, which were dark brown. There were a large number of inflammatory cells infiltration in the lamina propria, accompanied by vasodilatation and prolonged balloon discharge. Anorectal resting pressure showed decreased, the percentage of open arm time(OT%) and the percentage of open arm entry(OE%) were decreased. The content of brain-gut peptide in serum colon tissue were abnormal. And there was no significant change in gastrointestinal transit function. Conclusion Low-fiber diet combined with perianal area injection methylene blue on rats model presented the pathological characteristics of FDD, which could be an ideal composite animal model for the study of pathogenesis and pharmacotherapeutics.
