TMEM176A基因甲基化是胃癌诊断的分子标志物
TMEM176A methylation is a potential diagnostic marker of gastric cancer
【摘要】 目的研究TMEM176A基因在人胃癌中的甲基化情况及其临床意义。方法应用甲基化特异性PCR技术对209例胃癌组织进行分析。用癌症基因组图谱(TCGA)数据库进一步分析TMEM176A的表达受甲基化调控。结果 TCGA数据库分析发现,TMEM176A的表达与K450甲基化芯片结果显示的启动子区的18个CpG位点甲基化呈负相关(R=-0.622,P<0.0001)。甲基化特异性PCR检测发现,TMEM176A在胃癌中的甲基化率为68.42%(143/209),TMEM176A甲基化与淋巴结转移相关(P<0.05)。结论 TMEM176A在胃癌中频繁发生甲基化,TMEM176A基因的表达受启动子区甲基化的调控,TMEM176A基因是潜在的胃癌诊断和预后标志物。
【Abstract】 Objective To explore the methylation status and clinical values of TMEM176 A gene in human gastric cancer.Methods The methylation status and mRNA expression levels of TMEM176 A were extracted from TCGA database. 209 cases of gastric cancer were detected by methylation specific PCR(MSP).Results The expression of TMEM176 A was associated with methylation of 18 CpG sites in the promoter region reversely(R=-0.622, P<0.0001). The methylation status of TMEM176 A was detected by MSP in 209 cases of human gastric cancer. Methylation of TMEM176 A was found in 68.42%(143/209) of human gastric cancer samples. Methylation of TMEM176 A was significantly associated with lymph node metastasis(P<0.05).Conclusion TMEM176 A is frequently methylated in human gastric cancer, methylation of TMEM176 A is a potential gastric cancer diagnostic marker.
