咽喉反流性疾病患者肠道菌群变化的初步研究
Intestinal Microbiota Analysis of Patient with Laryngopharyngeal Reflux Disease
【摘要】 目的观察咽喉反流性疾病(laryngopharyngeal reflux disease,LPRD)患者与健康人肠道菌群组成的差异。方法选取LPRD患者30例为LPRD组,健康志愿者30例为对照组,采集两组对象新鲜粪便标本,利用16S rDNA高通量测序技术对两组粪便细菌群落组成结构及多样性进行分析。结果两组间alpha多样性和beta多样性分析均无显著性差异(P>0.05),LEfSe差异分析显示,LPRD组与对照组相比,放线菌门(P=0.030)、红蝽菌纲(P=0.010)、红蝽菌目(P=0.010)、生丝菌科(P=0.022)、红蝽菌科(P=0.010)、海洋杆菌属(P=0.022)、柯林斯氏菌属(P=0.004)、瘤胃球菌属(P=0.008)、普雷沃氏菌种(P=0.019)相对丰度有显著差异,其中放线菌门、红蝽菌纲、红蝽菌目、红蝽菌科、柯林斯氏菌属、瘤胃球菌属、普雷沃氏菌种在LPRD组富集,生丝菌科、海洋杆菌属在对照组富集。结论 LPRD患者与健康人肠道菌群多样性和物种组成无明显差异,但在菌群结构上发生了变化,一些特定细菌类群比例有差异,这些结构差异菌可能与LPRD的发生和发展有关。
【Abstract】 Objective To compare the intestinal microbiota between patients with laryngopharyngeal reflux disease(LPRD) and healthy people.Methods From Jan 2019 to May 2019,30 patients with LPRD in the ENT clinic of the Eighth Medical Center of Chinese PLA General Hospital were enrolled into the LPRD group, while 30 healthy people were included as control group.The feces were collected and total fecal DNA was extracted, and then all the microorganisms in the intestinal tract of LPRD patients and healthy people were detected and analyzed by 16 S rDNA-based high throughput sequencing for operational taxonomic units(OTUs) clustering.Species information analysis was performed according to the sequencing data in order to obtain the corresponding species information and abundance distribution.Results The alpha diversity and beta diversity of the intestinal microbiota had no significant difference between the LPRD group and the control group(P>0.05). Linear discriminant effect size(LEfSe) analysis revealed that the bacterial taxa with significantly different relative abundances in the LPRD group were actinobacteria, coriobacteriia, coriobacteriales, coriobacteriaceae, collinsella, ruminococcus, prevotella_sp__DJF_LS16,hyphomicrobiaceae and pelagibacterium compared with the control group.Conclusion There were no significant differences in the diversity and species composition of intestinal flora between LPRD patients and healthy people, but there were some changes in the bacterial structure, which showed the differences in the proportion of some specific bacterial groups. These structural differences may be related to the occurrence and development of LPRD.
