杜仲内生细菌的分离鉴定及活性检测

【摘要】 目的从杜仲Eucommia ulmoides的树皮、叶片和果实中分离出具有活性的内生细菌。方法采用平板培养法分离内生细菌,扩增16S rDNA序列并测序,在GenBank上Blast鉴定其归属;通过菌丝速率生长法和纸片琼脂扩散法检测抑菌活性,通过T-AOC试剂盒检测抗氧化活性。结果获得内生细菌37株,其中21株鉴定到属,分属于不动杆菌属(Acinetobacter sp.)、粪产碱菌属(Alcaligenes sp.)、葡萄球菌属(Staphylococcus sp.)和芽孢杆菌属(Bacillus sp.)。8株内生细菌具有抑菌活性,11株内生细菌具有抗氧化活性。EU1、EU3和EU12至少对4种病原菌具有抑制效果,其中EU1(芽孢杆菌属Bacillus sp.)对蜡样芽胞杆菌的抑制效果最好,其抑制率高达89.32%。EU3具有更好的抗氧化能力。结论分离到的一些内生细菌可应用于农业和天然药物的研究开发。

【Abstract】 Objective To isolate the active endophytic bacteria from the bark, leaves and fruits of Eucommia ulmoides. Methods Endophytic bacteria were isolated by plate culture method, the 16 S rDNA sequence was amplified and sequenced, the attribution was identified by Blast at GenBank,the antibacterial activity was detected by mycelium growth rate method and paper agar diffusion method, and the antioxidant activity was detected by T-AOC kit.Results A total of 37 endophytic bacteria were isolated and 21 were identified, belonging to Acinetobacter sp., Alcaligenes sp., Staphylococcus sp., and Bacillus sp.. Eight strains of endophytic bacteria have antibacterial activity, and 11 strains of endophytic bacteria have antioxidant activity. EU1, EU3 and EU12 have inhibitory effects on at least 4 pathogenic bacteria, among which EU1(Bacillus sp.) has the best inhibitory effect on Bacillus cereus, with its inhibitory rate as high as 89.32%. EU3 has better antioxidant capacity. Conclusion Some endophytic bacteria isolated can be used in agricultural and natural medicine research and development.
