生物纳米材料透射电镜样品制备条件研究
Study on the Preparation Conditions of Transmission Electron Microscope Sample of Biological Nanomaterials
【摘要】 在拍摄生物纳米材料透射电镜照片时,由于组成元素原子序数低,需要通过染色来增加图像衬度。从染液的选择、支持膜的处理、样品浓度、染色时间着手,通过实验分析了相关条件对染色后TEM图像效果的影响,从而为调节染色参数提供参考。选择适合的染液,才能获得理想的图像;对支持膜进行亲水性处理,可以减少由于界面纳米气泡而产生的球形假象;调节适宜的样品浓度,才能获得合适的样品分布;合适的染色时间,才能获得足够的染色衬度以及清晰的轮廓。
【Abstract】 Due to the low atomic numbers of the components of biological nanomaterials, it is necessary to increase the contrast of images by negative staining before taking transmission electron microscope photos. In this paper, the factors that might affect the TEM image effects, such as the stain type, the pretreatment of supporting membrane, the sample concentration and the staining time were investigated. Some possible phenomena under inappropriate conditions were studied, so as to provide a reference for the optimization of staining conditons, to obtain high quality TEM photos with clear boundary and obvious contrast. Choosing the right stain can get the ideal image; Hydrophilic treatment of the supporting membrane can reduce the spherical illusion caused by interfacial nanobubbles; proper sample concentration can get suitable sample distribution; proper staining time is necessary to obtain sufficient contrast and clear contour.
