粗毛纤孔菌发酵液中抗肿瘤代谢产物的分离与鉴定

【摘要】 粗毛纤孔菌(Inonotus hispidus)经液体发酵后,通过萃取、硅胶柱层析、LH20葡聚糖凝胶柱层析及高效液相色谱对粗毛纤孔菌的发酵液进行分离纯化;通过质谱、核磁共振技术分析鉴定化合物的结构。共分离并鉴定到麦角甾醇(1)、Hispolon(2)、肉桂酸(3)、尿苷(4)、Cyclo (L-Leu-L-Phe)(5)5个具有抑制Hela细胞活性的化合物,其中化合物3、4、5首次从粗毛纤孔菌中分离,化合物5还未有抗肿瘤活性方面的报道。MTT实验结果表明,化合物5能明显抑制Hela细胞,其半抑制浓度(IC50)为21μg·mL-1,且利用流式细胞术分析发现化合物5可促使Hela细胞发生凋亡。研究结果可为粗毛纤孔菌发酵液中抗肿瘤活性代谢产物的开发利用提供参考。

【Abstract】 Fermentation broth of Inonotus hispidus was isolated and purified by extraction, silica gel column chromatography, LH20 dextran gel column chromatography and high performance liquid chromatography. Then the isolated compounds were identified by mass spectrometry and nuclear magnetic resonance. The results showed that five compounds were found to have inhibitory activity against Hela cells and they were identified as follows: ergosterol(1), Hispolon(2), cinnamic acid(3), uridine(4) and Cyclo(L-Leu-L-Phe)(5). Among them, compounds 3, 4 and 5 were isolated from I. hispidus for the first time, and compound 5 has not been reported yet in terms of antitumor activity. The result of MTT assay showed that compound 5 significantly inhibited Hela cells with an IC50 of 21 μg·mL-1; and the result of flow cytometry analysis showed that compound 5 promoted cell apoptosis in Hela cells. This study provided an important reference for development and utilization of antitumor metabolites in I. hispidus.
