鸦胆子苦醇对三阴性乳腺癌细胞凋亡的影响
Effects of BRU on apoptosis of triple negative breast cancer cells
【摘要】 目的:研究鸦胆子苦醇(BRU)对三阴性乳腺癌MDA-MB-231细胞的凋亡影响。方法:用BRU处理MDA-MB-231细胞。采用CCK-8法检测MDA-MB-231细胞生长抑制率,流式细胞仪检测细胞的凋亡率和周期。结果:BRU对三阴性乳腺癌MDA-MB-231细胞生长抑制率测定结果显示,不同浓度的BRU对MDA-MB-231细胞生长抑制率与对照组相比存在明显差异,均具有统计学意义(均P<0.05),且呈时间与浓度依赖性,凋亡结果显示,经5、10、20、40μmol/L BRU作用细胞48 h后,随着浓度增高细胞的凋亡率也相应提高,与空白对照组相比差异均有统计学意义(均P<0.05)。周期结果提示经5、10、20、40μmol/L BRU作用细胞48 h后,随药物浓度增加,处于G0/G1期的细胞比例逐渐增加,S/G2/M期的细胞比例下降,与空白对照组相比差异均有统计学意义(均P<0.05)。结论:中药鸦胆子苦醇在体外能抑制三阴性乳腺癌MDA-MB-231细胞的生长,并能诱导该细胞凋亡。
【Abstract】 Objective:To study the effect of BRU on apoptosis of MDA-MB-231 cells in triple negative breast cancer.Methods:MDA-MB-231 cells were treated with BRU.CCK-8 method was used to detect the growth inhibition rate of MDA-MB-231 cells.The apoptosis rate and cell cycle were detected by flow cytometry.Results:The results of the determination of growth inhibition rate of MDA-MB-231 cells in triple negative breast cancer by BRU showed that the growth inhibition rate of MDA-MB-231 cells in different concentrations of BRU was significantly different from that in the control group(all P<0.05),which was time-dependent and concentration dependent.Apoptosis results showed that the cells were treated with 5,10,20 and 40 μmol/L BRU for 48 hours, the apoptosis rate of the cells increased with the increase of the concentration, and the differences were statistically significant compared with the blank control group(all P<0.05).Cell cycle results suggested that the cells were treated with 5,10,20,40 μmol/L BRU for 48 h, the proportion of cells in G0/G1phase gradually increased with the increase of drug concentration, and the proportion of cells in S/G2/M phase decreased, with statistically significant differences compared with the blank control group(all P<0.05).Conclusion:Brusatol can inhibit the growth of MDA-MB-231 cells in triple negative breast cancer and induce their apoptosis in vitro.
