基于Oncomine和GEPIA数据库分析NFE2L3基因在胃癌组织中的表达及其临床意义

【摘要】 目的:通过数据库挖掘分析NFE2L3基因在胃癌组织中的表达水平及其临床意义。方法:通过Oncomine数据库和GEPIA网站分析NFE2L3基因在胃癌组织中的差异性表达及其与生存预后关系;胃癌与正常组织的表达采用t检验分析,采用单因素方差分析胃癌不同临床分期的表达差异,使用Pearson指数分析胃癌相关研究,使用Kaplan-Meier数据分析胃癌患者生存及预后情况,并通过String数据库分析NFE2L3基因上下游的蛋白相互作用及调控关系。结果:胃癌组织中NFE2L3 mRNA表达水平显著高于胃正常组织;GEPIA分析发现高表达的NFE2L3基因与错配修复基因MSH2、MSH6、PSM2有关(P<0.05),与错配修复基因PMLH1无明显意义(P>0.05),NFE2L3与胃癌临床分期无相关性,但是高表达的NFE2L3基因与低生存率及不良预后有关(P<0.05)。蛋白质分析网络显示NFE2L3可能与HCFC1、OSBPL3、OSBPL6、HNRNPA3、NFE2L2、 MAFG、MAFF、MAFK、BACH1、NPRL3等蛋白相互作用。结论:通过多个数据库研究发现NFE2L3在胃癌组织中呈高表达、表达水平与胃癌患者生存与预后有关;NFE2L3可作为与胃癌预后相关的生物标志物及治疗靶点,参与肿瘤发生发展。

【Abstract】 Objective:To analyze the expression level and significance of NFE2 L3 gene in gastric cancer through database mining.Methods:The Onconmine database and GEPIA website were used to analyze the differential expression of NFE2 L3 gene in gastric cancer and the relationship between survival and prognosis.The expression of gastric cancer and normal tissues were analyzed by t-test,and one-way variance analysis was used to analyze the expression differences of different clinical analyses of gastric cancer,using Pearson index to analyze related research,use Kaplan-Meier database to analyze the survival and prognosis of gastric cancer patients,and use String database to analyze the protein interactions and regulatory relationships between the upstream and downstream of NFE2 L3 gene.Results:The expression level of NFE2 L3 mRNA in gastric cancer tissues in the control group was significantly higher than that in normal gastric tissues.GEPIA analysis revealed that the highly expressed NFE2 L3 gene was related to the mismatch repair genes MSH2 and MSH6(P<0.05),and was related to the mismatch repair genes PSM2 and MLH1 no significant significance(P>0.05).NFE2 L3 had no correlation with the clinical staging of gastric cancer,but high expression of NFE2 L3 was related to poor survival and prognosis(P<0.05).The protein analysis network showed that NFE2 L3 may interact with proteins such as HCFC1,OSBPL3,OSBPL6,HNRNPA3,NFE2 L2,MAFG,MAFF,MAFK,BACH1,NPRL3.Conclusion:Through multiple database studies,it is found that NFE2 L3 is highly expressed in gastric cancer tissues,and the expression level is related to the survival and prognosis of gastric cancer patients.NFE2 L3 can be used as a biomarker and treatment target related to the prognosis of gastric cancer,and participate in the development of tumors.
