上调SCD1对乳腺癌细胞增殖凋亡的影响及与其相关通路的调控机制

【摘要】 目的:探究上调硬脂酰辅酶A去饱和酶1(SCD1)对乳腺癌细胞增殖凋亡的影响及与其相关通路的调控机制。方法:将细胞分为三组,即对照组、阴性对照组和SCD1组。检测和比较每组的转染效率、细胞活力、增殖情况以及凋亡率。qPCR检测mRNA水平,Western blot检测蛋白表达水平。结果:SCD1组细胞的SCD1 mRNA和蛋白的表达水平显著高于对照组和阴性对照组。SCD1组的细胞活力和克隆形成数目显著高于对照组和阴性对照组。SCD1组的细胞凋亡率显著低于对照组和阴性对照组。转染SCD1后,细胞表达的Cyclin D1、PCNA和Bcl-2 mRNA水平显著升高,Caspase3、Bax mRNA水平显著降低。乳腺癌MCF-7细胞SCD1过表达后,Wnt、β-catenin mRNA和蛋白的表达水平均显著升高。结论:上调SCD1的水平可通过促进Wnt/β-catenin通路促进乳腺癌细胞的增殖并抑制其凋亡。

【Abstract】 Objective:To investigate the effect of stearoyl-CoA desaturase 1(SCD1) on proliferation and apoptosis of breast cancer cells and the regulation mechanism of related pathways.Methods:The cells were divided into three groups, including the control group, the negative control group and the SCD1 group.The transfection efficiency, cell viability, proliferation, and apoptotic rate of each group were detected and compared.qPCR detected mRNA level and Western blot detected protein expression level.Results:The expression level of SCD1 mRNA and protein in the SCD1 group were significantly higher than those in the control group and the negative control group.The cell viability and the number of clone formation in the SCD1 group were significantly higher than that in the control group and the negative control group.The apoptotic rate of the SCD1 group was significantly lower than that of the control group and the negative control group.After transfection of SCD1,the expression level of Cyclin D1,PCNA and Bcl-2 mRNA was significantly increased, and the mRNA level of Caspase3 and Bax was significantly decreased.After overexpression of SCD1 in breast cancer MCF-7 cells, the expression level of Wnt and β-catenin mRNA and protein was significantly increased.Conclusion:Up-regulation of SCD1 level promotes proliferation and inhibits apoptosis of breast cancer cells by promoting the Wnt/β-catenin pathway.
